• Note (General): type:text; 加齢に伴う個体レベルでの免疫系の機能的変化、いわゆる「免疫老化」は病原菌に対する易感染、自己免疫疾患や発がんと密接に関わっている。免疫老化は、主にCD4 T細胞を介した獲得免疫の異常によるものと考えられているが、とりわけ加齢に伴い増加するメモリーCD4 T細胞（免疫老化関連CD4 T細胞）の発生機序はほとんど明らかにされていない。また、タンパク質翻訳後修飾の一つであるユビキチン修飾も、免疫老化のプロセスにおけるその関与は不明である。そこで本研究では、CD4 T細胞を介した免疫老化の制御メカニズムの解明を目指し、ユビキチン関連分子及び転写因子やそれらを標的としたshRNA発現ライブラリーを導入したCD4 T細胞を用い、in vivo における免疫老化関連CD4 T細胞の発生に関わる因子の探索を実施することで、 以下の成果を得た。まず、110週齢を超えた老齢マウスの脾臓において、TCR刺激低応答性かつ炎症性素因の発現を示す、PD-1陽性の免疫老化関連CD4 T細胞の顕著な蓄積を見出した。また、RNAシーケンスによる遺伝子発現解析を行った結果、老齢マウス由来のメモリーCD4 T細胞では、細胞傷害性（cytotoxity）や疲弊（exhaustion）に関与する遺伝子の発現上昇が認められた。さらに、このような免疫老化関連CD4 T細胞は、CD4 T細胞をT細胞欠損マウスへ移入することによって、短期間で実験的に誘導することが可能であった 。そこで、発現ライブラリーを導入したCD4 T細胞をRAG2欠損マウスに移入し、cytotoxityやexhaustionを制御する遺伝子の探索を行った。その結果、52個の探索遺伝子のうち3個をcytotoxity、5個をexhaustionの制御因子の候補としてそれぞれ同定した。興味深いことに、これらの遺伝子はいずれも免疫老化における関与が明らかにされていないものであった。今後、本研究をさらに発展させ、同定因子による新規免疫老化制御機構の詳細を明らかにすることで、高齢者における獲得免疫能の改善のための予防、治療法の開発に貢献することを目指す。 The age-associated decline of immune functions, so-called immunosenescence is featured with increased susceptibility of infection, autoimmunity and tumor incidence. Although accumulated memory CD4 T cell during aging might play an important role in immunosenescence, detailed mechanisms are largely unknown. Protein ubiquitination, which is one of the posttranslational modifications, is involved in T cell activation and differentiation, whereas it's not clear whether this regulates immunosenescence. To understand the cellular and molecular mechanisms how senescence-related CD4 T cells are emerged and control immunosenescence, we tried to identify the master regulator that functions in this process, using the in vivo screening system based on transduction of retroviral library which includes genes encoding ubiquitin-related molecules and transcription factors. Firstly we found that senescence-related CD4 T cells that are defined as PD-1+CD44+ CD4 T cells were prominent in aged mice. RNA seq revealed that accumulated memory CD4 T cells in aged mice acquired cytotoxic and exhausted properties. Furthermore, such senescence-related CD4 T cells were experimentally induced in CD4 T cells that were adaptively transferred into lymphopenic mice. Thus we transferred library-transduced CD4 T cell into RAG2 deficient mice and assessed cytotoxic and exhausted CD4 T cells in the spleen. Among tested 52 genes, 3 genes related to cytotoxic and 5 genes related to exhausted CD4 T cell were identified as the candidates, respectively. We are further investigating whether and how those molecules function in CD4 T cell mediated immunosenecence. This may provide us not only molecular insight of immunosenescence, but also a new strategy for prevention and treatment of age-related diseases.
