• Note (General): 出版タイプ： VoR; type:text; 本研究では生体外で細胞を三次元的に培養するための足場材料として，高静水圧を用いた脱細胞化組織の作製することを目的としている．最終的には，脱細胞化組織を用いた肝・腎組織の再生医療への適用を目指している．本年度は，油圧駆動型の材料試験機に設置可能で，かつ，250 MPaの高静水圧に耐えうる強度の脱細胞化処理用の耐圧容器を開発した．さらに，細胞懸濁液，生体組織（臓器）の模擬体として細胞含有ハイドロゲルを対象とした高静水圧の印加による脱細胞化実験を実施した．その結果，細胞懸濁液を対象とした試験では250 MPaの高静水圧を印加することで80％以上の細胞死を誘導し，さらに細胞から核が除去される脱核が実現できることが明らかとなった．同じく細胞包含ハイドロゲルにおいても250 MPaの高静水圧の印加によって細胞死の誘導および脱核が実現できた．今後はラットなどの動物組織を対象として高静水圧を用いた脱細胞化組織の作製を実施し，高静水圧の印加条件が細胞死，脱核に与える影響を明らかにする． The purpose of this study is to produce decellularized tissue using high hydrostatic pressure as a scaffold for in vitro three-dimensional cell culture. Final goal of this study is to apply this decellularized material for liver and kidney tissue regeneration. This year, we developed a pressure-resistant chamber that can be installed on a material testing machine and that can bear high hydrostatic pressure of 250 MPa. In addition, decellularization experiments were performed by imposing high hydrostatic pressure on cell suspension and cell-containing hydrogel simulating a living tissue. As a result, application of a high hydrostatic pressure of 250 MPa induced more than 80% cell death in both cell suspension and cell-containing hydrogel, and further achieved enucleation. In the future, we will develop a decellularized material by imposing a high hydrostatic pressure on animal organs and clarify the effects of high hydrostatic pressure on cell death and enucleation.
