• Note (General): type:text; 本研究プロジェクトの目的は、薬物試験や再生移植医療のための成熟したヒト心筋組織をin vitroで構築する技術の確立である。世界に先駆けin vitroマイクロファイバー組織を開発した本塾理工学部機械工学科の尾上グループ（尾上）と、多能性幹細胞由来の再生心筋細胞の純化精製法において世界トップクラスの技術を有する本塾医学部循環器内科の福田グループ（福田，藤田，遠山）との医工連携により研究開発を実施した。 ３年計画の最終年度である本年は、組織の成熟化のため長期培養が可能となる培養条件の探索を行った。昨年度判明した課題として、長期間ファイバ内にて高純度化された心筋細胞を培養すると、次第に断片化が進行することが挙げられた。そこで本年度は、ヒトiPS由来心筋細胞への線維芽細胞の添加、線維芽細胞増殖因子の培地への添加、およびファイバのシェル層であるアルギン酸ゲルの除去などを行い、安定化の条件の検討を行った。これにより、細胞数が重要なパラメータであることが見いだされた。またこの検討の中で、ファイバ内への共培養技術の展開により血管内皮細胞を混合した細胞ファイバによる組織構築技術が完成した。その成果は国際学術誌APL Bioengineeringに採択され表紙および2019年度のハイライト論文に選出された。 今後は本研究プロジェクトの最終目的である成熟したin vitro組織構築のため、力学刺激装置の開発とさらなる培養条件の検討を進める予定である。 The aim of this project is to establish technologies to construct in vitro matured induced pluripotent stem (iPSC)-derived human cardiac tissues for drug development and regenerative medicine. This project was launched by the collaboration between Onoe group in Mechanical Engineering and Fukuda group in Cardiology. In this year (the last year in three-year term project), we have explored the optimal condition to maintain fiber-shaped human iPS cell-derived cardiac muscle tissue for long term to mature the tissue. Last year, we encountered the problem that the iPS cell-derived cardiac tissues tended to shrink in the microfiber-shaped hydrogel scaffold in long-term culture (over 2 weeks). To solve this, we tested three different culture conditions: (i) addition of fibroblast to iPS cell-derived cardiac cell to stabilize the tissue shape, (ii) addition of fibroblast growth factor to the culture medium of the microfiber-shaped iPS cell-derived cardiomyocyte tissues, and (iii) removal of the alginate shell of the microfiber-shaped cardiac tissue to promote the exchange of nutrients and wastes. As a result, we found that the cell number in the hydrogel microfiber was the one of the important parameters to maintain the microfiber-shaped tissue for long term. Through these experiments, we have developed a co-culture method for the microfiber-shaped tissues where endothelial cells were mixed for building macroscopic assembled tissues. This achievement was accepted in the international journal APL Bioengineering and selected as a cover image and selected to the highlighted article in 2019. Toward next stage, we are focusing on developing tissue culture systems with controlled mechanical stimulus for constructing matured iPS cell-derived artificial tissues.
