大腸菌に組換え体として外来タンパク質を強制発現させる場合、非常によく発現するタンパク質と、そうではないタンパク質に大きく2分される。本申請研究の最終目的は、組換え体の産生が極めて厳しいようなタンパク質に関して、その物質生産に歯止めをかけていたような大腸菌側の要因を取り除き、効率の良い外来タンパク質の発現系を構築することにある。また、これを、人工RNAシステムを用いることで可能にする。これまでの研究の成果として、超好熱性アーキアであるパイロコッカス・フリオサスに由来するPF1614タンパク質 (潜在的DNA/RNA結合タンパク質)は大腸菌に組換え体として発現させることが困難なことが (未発表)、一方で、PF0027タンパク質 (金井ら2009, RNA)は非常によく発現することが分かっている。そこで、大腸菌にPF1614タンパク質やPF0027タンパク質を強制発現させるような条件下において、PF1614タンパク質の発現に依存して、増減する大腸菌由来のタンパク質を、ショットガンプロテオミクス法にて解析した。結果、約1800種のタンパク質が定量的に同定され、PF0027タンパク質の発現誘導では、数百の大腸菌タンパク質に発現の増減が見られるが、PF1614タンパク質の場合では、わずか数十のタンパク質に絞られることが分かった。PF1614タンパク質の誘導に依存して発現が上昇するタンパク質、すなわちPF1614(+)/PF1614(-) の発現変動比 >1.4 には、タンパク質の細菌外膜への挿入や折りたたみを行っているBAM（β-barrel assembly machinery）複合体因子や、LPS-アッセンブリータンパク質、内膜に存在するタンパク質群、特定のリボソームタンパク質等があった。また、PF1614タンパク質の誘導に依存して発現が下降するタンパク質、すなわち、PF1614(+)/PF1614(-) の発現変動比＜0.71には、ペリプラズムシャペロンであるspyや特定のリボソームタンパク質等があった。今後、このうちのどのタンパク質が実際にPF1614の発現を制御しているのかを解析していく予定である。 The goal of this research is to construct an efficient recombinant protein expression system in Escherichia coli by removing factors that inhibit the production of the recombinant protein. We plan to make it possible using an artificial RNA system. As the first step, we conducted a whole proteome analysis of proteins extracted from E. coli overexpressing the PF1614 protein that is an archaeal protein hardly produced in E. coli as a recombinant protein. The proteome analysis detected approximately 1,800 E. coli proteins and we found dozens of E. coli proteins whose expression change depending on the induction of PF1614 protein. Many of them were membrane-related proteins. In the future, we plan to analyze which of these proteins actually controls PF1614 production.
