第一膜貫通領域の23アミノ酸上流までのα1nAChR細胞外領域（α1nAChRex）をPGGGGSリンカーでOKTに接続したα1nAChRex-OKTを恒常的に発現するRK13細胞を樹立した。得られたα1nAChRex-OKTタンパク質を、抗α1抗体（mAb35）を産生するハイブリドーマTIB-175細胞、ヒトCD3を発現するJurkat細胞を用いてフローサイトメトリーでの結合性の評価を行った。Jurkat細胞との結合は確認できたが、TIB-175細胞との結合は確認できなかった。TIB-175細胞が形質膜上にラット抗体を発現していることは、ラット抗体の軽鎖を認識する抗体（MRK-81）を用いたフローサイトメトリーによる解析で確認しているため、TIB-175細胞との結合が確認できなかった一因として、α1nAChRex-OKTタンパク質のタンパク質濃度が低いことが考えられた。そこで、α1nAChRex-OKTタンパク質の産生効率などを改善すべく、リンカー長の変更やα1nAChRex構造の変更を行った。α1nAChRex構造の変更に関しては、主要免疫原性領域を含むα1nAChRexの最小化や、分子構造に影響を与えると考えらえる配列の変異導入、nAChRが5量体を形成することから、抗体の重鎖定常域1（constant heavy 1 : CH1）と軽鎖定常域（constant light : CL）を用いた2量体化などを試みた。結果、主要免疫原性領域を含む最小配列で、2つのエピトープの中間部位に変異を導入したα1nAChRexをDKTHTGSリンカーでOKTに接続した融合タンパク質（α1nAChRex改変-OKT）で培養上清への高いタンパク産生と、Jurkat細胞への結合を確認することができた。次に、タンパク質医薬品産生に多用されているCHO DG44細胞を用いて、α1nAChRex改変-OKTを安定的に産生される細胞を樹立した。この細胞の培養上清より精製したα1nAChRex改変-OKTを用いて、TIB-175細胞への結合をフローサイトメトリーにより確認した結果、結合を確認するに至らなかった。今回、作製した主要免疫原性領域を含むα1nAChRex改変-OKTタンパクでは、タンパク質濃度による問題では無く、mAb35により認識されない可能性が高いと考えられる。既に実用化が検討されているα1nAChRex-Fcに関しては、TIB-175細胞への結合と細胞障害活性が確認されている。抗体によるエピトープ認識には立体構造が大きく影響しているため、更なるα1nAChRex配列の改変を進める予定である。また、細胞障害活性の比較、TIB-175細胞への結合の参考のためα1nAChRex-Fcタンパク質の精製も合わせて行なう予定である。 RK13 cells were established that constantly express α1nAChRex-OKT, in which the α1nAChR extracellular region up to 23 amino acids upstream of the first transmembrane region (α1nAChRex) is connected to OKT with a PGGGS linker. The resulting α1nAChRex-OKT protein was evaluated for binding by flow cytometry using hybridoma TIB-175 cells producing anti-α1 antibody (mAb35) and Jurkat cells expressing human CD3. binding to Jurkat cells was confirmed, but binding to TIB-175 Since the fact that TIB-175 cells express rat antibodies on their plasma membrane was confirmed by flow cytometry analysis using an antibody that recognizes the light chain of rat antibodies (MRK-81), one reason for the failure to confirm binding to TIB-175 cells was that low protein concentration of α1nAChRex-OKT protein was considered. To improve the production efficiency of the α1nAChRex-OKT protein, the linker length and the α1nAChRex structure were changed. Mutations were introduced, and since nAChR forms a pentamer, dimerization using the constant heavy 1 (CH1) and constant light (CL) regions of the antibody was attempted. As a result, we confirmed high protein production in culture supernatants and binding to Jurkat cells by a fusion protein (α1nAChRex-modified-OKT) in which α1nAChRex, a minimal sequence containing the major immunogenic region and a mutation in the intermediate site of two epitopes, was connected to OKT with a DKTHTGS linker. Next, using CHO DG44 cells, which are frequently used for protein drug production, we established cells that stably produce α1nAChRex-modified-OKT. The α1nAChRex-modified-OKT purified from the culture supernatant of these cells was confirmed by flow cytometry for binding to TIB-175 cells, but binding was not confirmed. The α1nAChRex-modified-OKT protein containing the major immunogenic region prepared in this study is not likely to be recognized by mAb35, not because of a problem with the protein concentration. As for α1nAChRex-Fc, which is already being considered for commercialization, binding to TIB-175 cells and cytotoxic activity have been confirmed. Since the three-dimensional structure has a significant effect on epitope recognition by antibodies, further modification of the α1nAChRex sequence is planned. We also plan to purify α1nAChRex-Fc protein for comparison of cytotoxic activity and binding to TIB-175 cells.
