• Note (General): type:text; 子宮ラセン動脈は子宮内膜脱落膜化に従い形成され、妊娠が成立した場合には妊娠初期に再構築されることで、交感神経支配から回避と血管系を拡張し、妊娠中期以降の胎児胎盤系への血液供給に重要な役割を果たす。これまでに子宮ラセン動脈再構築不全は胎盤虚血や妊娠高血圧症候群、胎児発育不全を惹起することが指摘されている。子宮ラセン動脈再構築には胎児由来細胞である血管内絨毛外栄養膜細胞（enEVT）が母体の血管内皮細胞と置換される機構が重要である。本研究においては、enEVTモデルとしてHTR-8細胞、血管内皮細胞としてHUVEC細胞をそれぞれ用い、HTR-8細胞がHUVEC細胞に対する指向性浸潤を検討した。 HTR-8細胞を細胞外基質被覆した培養ウェルにおいて、陽性対照と位置付けた血管内皮細胞増殖因子添加時の浸潤能を示すことができた。HUVEC由来のenEVT浸潤誘導因子を探索するため、HUVECとHTR-8共培養系におけるHTR-8の浸潤を評価した。HUVECには未分化状態および血管形態のそれぞれの状態があるため、血管形態への分化がHTR-8の浸潤に及ぼす影響を評価した。予備検討結果においては、血管形態のHUVECにおいて、HTR-8の浸潤が抑制される現象が観察された。これは、成熟した内皮細胞においてはHTR-8の浸潤を誘導しない作用と理解され、enEVTが内皮細胞へ浸潤する因子を、内皮細胞自身が分泌していると推察された。 Uterine spiral arteries are formed by endometrial decidualization, and when pregnancy is established, they are reconstructed in the early stages of pregnancy to avoid sympathetic innervation and dilate the vascular system. It plays an important role in supplying blood to the placenta. It has been pointed out that uterine spiral artery remodeling failure causes placental ischemia, preeclampsia, and fetal growth restriction. The mechanism by which fetal-derived cells, endovascular extravillous trophoblast cells (enEVT), are replaced with maternal vascular endothelial cells is important for uterine spiral artery remodeling. In this study, HTR-8 cells were used as an enEVT model and HUVEC cells were used as vascular endothelial cells, and directional invasion of HTR-8 cells into HUVEC cells was investigated. In the culture wells coated by an extracellular matrix, invasion of HTR-8 cells by the addition of vascular endothelial growth factor was shown, which was positioned as a positive control. In order to search for the enEVT invasion-inducing factor derived from HUVEC, the invasion of HTR-8 in the HUVEC and HTR-8 co-culture system was evaluated. Since HUVEC has an undifferentiated state and a tube-like morphology, the effect of differentiation into tube-like morphology on HTR-8 invasion was evaluated. In the preliminary study results, it was observed that the invasion of HTR-8 was suppressed in the tube-like morphology of HUVEC. These results imply that mature endothelial cells does not induce the invasion of HTR-8, and it was speculated that the endothelial cells themselves secreted the factor inducing enEVT invasion.; 申請種類 : 福澤基金研究補助
