呼吸器疾患における免疫細胞の老化の影響を調べるため、若齢マウスと高齢マウスの肺から採取した自然リンパ球のSingle cell RNA-seq解析を行った。その結果、すべての自然リンパ球が加齢とともに遺伝子発現が変化し、自然リンパ球のサブタイプにも変化が生じることが明らかになった。特に、2型自然リンパ球(ILC2)は加齢とともにIL-5の発現が低下する一方、アンフィレギュリンなどの発現は変化しないことが判明した。さらに、加齢によって特定のILC3が著明に増加し、この細胞が加齢に伴う好中球性炎症に寄与することが示唆された。現在、これらのSingle cell RNA-seq解析の結果をもとに、老化と呼吸器疾患の関連をマウスモデルを用いて検討を行っている。これらの結果はまだ論文化はできていないが、今回の知見をもとに自然リンパ球の多様性に関する総説を作成した。 To investigate the aging effects of innate lymphocytes on respiratory disease, single cell RNA-seq analysis of innate lymphocytes collected from the lungs of young and aged mice was performed. The results revealed that gene expression of all innate lymphocytes changes with aging. In particular, group 2 innate lymphoid cells (ILC2s) were found to have decreased expression of IL-5 with aging, while expression of amphiregulin and other genes remained unchanged. Furthermore, a specific subtype of ILC3s markedly increases with aging, suggesting that these cells contribute to aging-associated neutrophilic inflammation. Based on the results of these single cell RNA-seq analyses, the relationship between aging and respiratory diseases is currently being investigated in various mouse models. Although these results have not yet been published, we have prepared a review article on the diversity of innate lymphocytes based on our findings.
