• Note (General): type:text; ヒト絨毛外栄養膜細胞（enEVTs） のモデル細胞株として、ヒト妊娠初期胎盤より単離され不死化細胞株として樹立されている HTR-8/SVneo（HTR-8）細胞を用いた。ヒト血管内皮細胞ののモデル細胞株としてHUEhT-1細胞を用いた。HUEhT-1細胞は、臍帯静脈由来内皮細胞株として汎用されるHUVEC細胞にteromeraseを遺伝子導入することにより不死化した細胞株である。HUEhT-1細胞は、培養条件により敷石状と管腔状の2形態をとる。HTR-8の浸潤能に対して、敷石状に培養したHUEhT-1細胞との共培養において、HTR-8の浸潤は促進されたのに対し、管腔状に培養したHUEhT-1細胞との共培養は促進を示さなかった。また、敷石状に培養したHUEhT-1細胞との共培養において、HTR-8細胞の遊走能および増殖能の促進は観察されなかった。これらの結果により、敷石状に培養したHUEhT-1細胞にHTR-8細胞の浸潤を誘導する因子が含まれる可能性が高まった。 HUEhT-1細胞によるHTR-8細胞の浸潤促進機構を解明するため、単培養のHTR-8および敷石状のHUEhT-1細胞と共培養したHTR-8細胞、敷石状HUEhT-1細胞、管腔状HUEhT-1細胞よりRNAを抽出し、RNAseq解析を行った。細胞の遊走あるいは浸潤を促進するケモカイン群およびその受容体群においては、HTR-8の浸潤促進を説明可能な組み合わせは見出されなかった。一方、HTR-8細胞のHUEhT-1細胞との共培養によるRNA発現変化により、細胞外マトリクスに変化を与える可能性が見出された。これは、上述の浸潤亢進と、非遊走亢進とも対応する結果であった。さらにRNA発現変動によりADAMファミリーおよびプラスミン発現パスウェイに変動が見られたことから、これらの因子がHTR-8の浸潤に影響を与えることが示唆された。 HTR-8/SVneo (HTR-8) cells, which were isolated from human placenta in early pregnancy and established as an immortalized cell line, were used as a model cell line for human extravillous trophoblasts (enEVT). HUEhT-1 cells were used as a model cell line for human vascular endothelial cells, which are immortalized by gene transfer of telomerase into HUVEC cells, a common endothelial cell line derived from umbilical veins. Co-culture with paving stone-shaped HUEhT-1 cells promoted HTR-8 invasion ability, whereas co-culture with tube-shaped HUEhT-1 cells did not. In addition, no enhancement of migration or proliferation capacity of HTR-8 cells was observed when co-cultured with paving stone-shaped HUEhT-1 cells. These results suggest that the paving stone-shaped HUEhT-1 cells may contain factors that induce HTR-8 cell invasion. To elucidate the mechanism by which HUEhT-1 cells promote HTR-8 cell invasion, total RNA was extracted from monocultured HTR-8 cells, paving stone-shaped HUEhT-1 cells, and HTR-8 cells co-cultured with tube-shaped HUEhT-1 cells, and RNAseq analysis was performed. As a result, we did not find any combination of chemokines and their receptors that promote cell migration or invasion that could explain the invasion-promoting ability of HTR-8. On the other hand, changes in RNA expression by co-culturing HTR-8 cells with HUEhT-1 cells may alter the extracellular matrix. These results are corresponded to the aforementioned promotion of invasion and non-migration. Furthermore, changes in ADAM family and plasmin expression pathways were observed, suggesting that these factors affect HTR-8 invasion.; 申請種類 : 福澤基金研究補助
