細胞レベルでの「老い」に伴ってクロマチン制御異常が起こるとされるが、このような「エピゲノム老化」が高次生命システムである免疫系細胞に起こるかは明らかになっていない。私たちは、老化個体の免疫細胞を解析し、ヒストンメチル化酵素と会合し、ヘテロクロマチン形成を維持するTRIM28 タンパクのセリン473残基のリン酸化が低下すること、それにともなって、クロマチン制御不全に伴う一連の遺伝子脱抑制が起こることを見出した。昨年までに、TRIM28を免疫細胞で欠損したマウスには、免疫不全、炎症形質、自己免疫疾患といった免疫老化の表現型があらわれる事を見出している。また、クロマチン免疫沈降シークエンス、RNAシークエンスの複合オミクス解析により、TRIM28欠損細胞では、ゲノム上で抑制を受けるべき内在性レトロウイルス配列（ERV）の脱抑制を起こすこと、免疫活性化や炎症に関わる多くの遺伝子には、特定のERV配列が近傍に存在し、ERVの脱抑制によって炎症性遺伝子が共に誘導され、炎症を起こしている事も明らかとなった。昨年までに、全身の細胞でTRIM28遺伝子を片方の染色体で欠損するヘテロマウス（TRIM28+/-）を作成し、このマウスが全身でTRIM28の低発現を起こすことから解析を行っている。結果としてTRIM28+/-マウスの一部が血中アミノ酸の低下やミトコンドリア面積の減少を伴う早期老化の表現型を示すことを見出した。RNAシークエンスの結果、興味深いことに、TRIM28欠損細胞では、がん抑制に深く関わるp53経路が活性化しており、TRIM28+/-においてはこれが全身に起こることで早期老化症状の原因となっている事が考えられた。今後は、TRIM28+/-における早期老化が、実際の組織老化と同様の組織学的変化を伴うのか、老化炎症形質を伴うのか、p53経路によるものなのか、という事を明らかにするため、このマウスの器官に起こる変化につき、組織学的解析を行っている。 It is suggested that cellular aging associates with aberrant gene expression caused by deregulation of chromatin. It is not clear if "epigenome aging" occurs on the immune system, upon aging, and affect its function. We previously reported that immune cells from old mice show reduction of TRIM28, a chromatin regulatory factor at phosphorylation level, associated with gene de-repression. Based on this observation, we generated immune cell-specific TRIM28 deficient mice, and found that these mice showed immunodeficiency, inflammation and autoimmunity, typical features of "immune-aging". Importantly TRIM28 deficient DCs showed aberrant expression of inflammatory genes that showed significant colocalization with particular endogenous retrovirus (ERV) elements on the genome, that contributed to expression of proximal inflammatory genes. To explore systemic effects of defective chromatin regulation, we generated mice with heterozygote deficiency of Trim28 gene (Trim28+/-), resulting in haploinsufficiency. We found that a fraction of Trim28+/- mice show growth defects with premature aging phenotype with defective mitochondria and low serum amino acids. Interestingly, Trim28 deficient cells showed high expression of p53, a tumor suppressor gene that may explain the aging phenotype. We are now working to elucidate the phenotype of Trim28+/- is reflecting real aging phenotype by examining histological changes that occur in these mice. In summary, our study suggested that alteration of epigenomic factor underlies epigenome aging, and can contributes to systemic phenotypes associate with natural aging.
