【背景および目的】我々が行った先行研究では，高脂肪食を摂取させた若齢肥満マウスの脂肪組織において老化関連遺伝子の発現が上昇し，PPARγアゴニストの投与はその作用を減弱化することが示唆されている．しかし老齢マウスの脂肪組織に対するPPARγアゴニスト投与の効果については未だ知見が少なく，不明な点が多く残されている．そこで本研究では，老齢マウスに対してPPARγアゴニスト(ピオグリタゾン)を投与し，脂肪組織における抗老化効果と寿命延長効果について検討を行った．【対象および方法】 88週齢の雄性C57BL/6マウスを対象に，普通食摂取群およびピオグリタゾン(0.02%,w/w)含有食摂取群を設けて12週間飼育した．100週齢時にサンプリングを行い，血中アディポネクチン濃度，脂肪組織の細胞サイズ，脂肪組織中の老化関連遺伝子の発現変化について解析を行った．また一部のマウスはピオグリタゾンの投与を継続し，死亡日までの日数の計測を行った．【結果および考察】老齢マウスに対するピオグリタゾンの投与によって，脂肪細胞サイズの縮小，血中アディポネクチン濃度の上昇，Mest遺伝子の発現低下を確認することができた．しかし一部の老化関連遺伝子およびSASP遺伝子の発現には，減少傾向がみられたものの有意な変化は認められなかった．また死亡日までの日数は現在まだ計測中だが，ピオグリタゾン投与によって中央値で約50日，平均値で約18日ほど延長する傾向がみられ，今後の検討結果が期待される．以上の結果から，老齢マウスに対するピオグリタゾン投与では，寿命の延長傾向と脂肪組織に対する細胞サイズの小型化誘導作用やアディポネクチンの分泌亢進作用は認められたものの，脂肪組織の老化様変化に対する改善効果は充分に確認できなかったことから，今後，継続的な寿命の観察とピオグリタゾンの投与量・投与期間などについての更なる検討が必要と思われた．【Background and Purpose】 In our previous study, pioglitazone ingestion could ameliorate senescence related markers in obese adipose tissue of young mice. The purpose of this study was to investigate the effects of pioglitazone (PPARγ agonist) on life longevity and senescence related markers in visceral adipose tissue of aged mice. 【Animals and Methods】 88-week-old male C57BL/6 mice were assigned to normal control diet (NCD) and pioglitazone ingestion (PGZ) groups. The mice in PGZ group were given 12-week dietary ingestion of 0.02 % (w/w) pioglitazone in normal control diet. At the end of experimental period, blood and abdominal fat tissues were collected for the analysis of plasma adiponectin concentration, adipocyte cellularity and senescence related markers. And we also recorded the life longevity of some mice in each group. 【Results and Discussions】 The significant increase in plasma adiponectin concentration, and significant decrease in averaged adipocyte sizes and MEST mRNA expression in adipose tissue were observed in PGZ group. However, no significant changes were observed in many senescence related markers and SASP mRNA expressions in adipose tissue of the PGZ groups with some exception. In the recording of life longevity, the PGZ group shows the extension of median (about 50-day) and average (about 18-day) life at this moment (still under experiment). These results suggested that the further experiments are needed about the effects on life longevity, and the effects of concentration, volume, and duration of the pioglitazone ingestion for the aged mice.
