• Note (General): type:text; C型レクチン受容体 (CLRs)の一種であるMincleは、主にマクロファージなどの免疫細胞表面に発現する受容体であり、結核菌の細胞壁構成成分である糖脂質トレハロースジミコール酸 (TDM)を初めとして、様々な生物種由来および自己の死細胞由来の複合脂質等をリガンドとして認識し免疫を制御する。これまでの研究から活性発現における脂質部位の重要性が明らかとなっているが、その構造活性相関の詳細は不明である。また、リガンドの極性部位 (トレハロース部位等)はMincleのカルシウムイオンおよびその周辺残基と相互作用することが示唆されているが、リガンド脂質部位とMincleの結合様式は不明である。さらに細胞表層上の受容体については、リガンド内在化等の細胞内挙動の報告例があるものの、Mincleのリガンド認識における細胞内挙動はほとんど解明されていない。 本研究では強力な Mincle アゴニストであり、特異な脂質構造を有するマンニトール複合脂質および、官能基修飾した脂質を含むトレハロースエステルの合成法を開発し、脂質改変誘導体を含む関連化合物ライブラリの構築および分子プローブ合成を行った。分子設計は事前の分子動力学(MD)計算より、Mincle 表層の極性アミノ酸残基を利用した。構築した化合物ライブラリに対する化合物評価を行うことで、Mincle の分子認識機構に関する新たな知見を得た。各複合脂質において、脂質部位構造の違いにおける活性の差異が見られた。またMincleのリガンド認識における細胞内挙動の観測を可能とするMincleリガンドを合成し、イメージング解析に展開することで細胞内挙動に関する知見を得た。以上の結果により、脂質部位構造の異なる誘導体を含むMincleリガンドのライブラリ構築に成功するとともにC型レクチン受容体Mincleの機能解明に成功した。 Mincle, a C-type lectin receptor (CLR), is expressed in activated phagocytes. The recognition of certain lipid conjugates from various organisms and dead host cells by Mincle leads to the activation of the innate immune system. The representative ligand is the Mycobacterium tuberculosis cell wall glycolipid trehalose dimycolate (TDM), and the other Mincle ligands are the lipid conjugate include glyco-, glycero-, and mannitol lipid conjugates. The importance of their lipid moieties for Mincle-mediated signaling activity have been demonstrated by the structure–activity relationship study of these ligand. The binding mode of polar head group of ligands (trehalose moiety) for Ca ion and its proximal site of Mincle was revealed, but the binding mode with lipid moiety and Mincle is still unclear. Furthermore, the internalization of Mincle by recognition of the ligand is not fully understood. In this work, we established the synthetic methods for mannitol lipid conjugates with the distinctive lipid structure and trehalose diester containing the lipids modified with polar groups, and to build their chemical library. The ligands were designed to interact the polar amino acid residue on the surface of Mincle utilizing MD simulation. We gained new insights for the molecular recognition of Mincle by the evaluation of their Mincle-mediated signaling activity. We also synthesized the fluorescence-labeled Mincle ligand and the cellular imaging studies were performed to the understanding of Mincle functions.
