PVニューロンの活動を特異的にイメージングするアデノ随伴ウイルス（AAV）ベクターを開発した。具体的にはてんかんの原因となっているScn1a遺伝子からPV特異的に発現するエンハンサーから作製された人工プロモーター（Schneider et al., Nat Neurosci. 2020）をもとにAAV1にパッケージングした（AAV-S5E2-GCaMP6f: 2.4×10^13 vg/ml）。同AAVベクターをラット運動野に注入し、内視鏡型蛍光顕微鏡を挿入したところ、ラット行動中の輝度値変化を検出することができた。また、同ベクターはマーモセットPVニューロンでGCaMPを発現することが組織学的に確認できた。マーモセットでカルシウムイメージングをおこなうためにはより強いGCaMPの発現量が必要になるため（Sadakene et al., Cell Rep. 2015; Kondo et al., Cell Rep. 2018）、次段階としてtetシステムにS5E2プロモーターを組み込む予定である。マーモセットにおける巧緻性の運動学習を行うためのマーモセット用のレバーを既に開発しており、2020年度はこれをマーモセットカルシウムイメージング用に改良を加えたところ、元の装置と同様のタイムコースで運動学習することが確認できた。本研究期間内にマーモセットPVニューロン活動のイメージングをおこなうための準備が整ったので、次年度はマーモセット運動学習過程におけるこれらの活動変化について解析を進めていく。 We have developed adeno-associated virus (AAV) vectors that specifically image the activity of PV neurons. We packaged AAV1 based on an artificial promoter (Schneider et al., Nat Neurosci. 2020) created from a PV-specific enhancer from the Scn1a gene, which is known to cause epilepsy (AAV-S5E2-GCaMP6f: 2. 4×10^13 vg/ml). The AAV vector was injected into the rat motor cortex, and an endoscopic fluorescence microscope was inserted to detect changes in luminance values during rat behavior. Histological analysis showed that the vector expressed GCaMP in marmoset PV neurons. Since stronger GCaMP expression is required for calcium imaging in marmosets (Sadakene et al., Cell Rep. 2015; Kondo et al., Cell Rep. 2018), we plan to incorporate the S5E2 promoter into the tet system as the next step. We have already developed a lever for marmosets to perform motor learning of elaboration in marmosets, and in FY2020, we modified it for marmoset calcium imaging and confirmed that it performs motor learning with the same time course as the original device. In the next year, we will analyze the changes in these activities during the marmoset motor learning process.
