前立腺がん患者は、ホルモン療法に抵抗性を持つ去勢抵抗性前立腺がん(CRPC)に進展することにより治癒困難となることが問題となっている。我々はこれまでに、がん幹細胞抗原であるCXorf48が前立腺がん幹細胞にmRNAレベルで高発現していることを確認している。さらに、CXorf48を特異的に認識するT細胞受容体(TCR)遺伝子のクローニングに成功した。本研究では、本TCR遺伝子をヒトＴ細胞株に遺伝子導入することでCXorf48特異的TCR-T細胞株を樹立し、前立腺がん細胞に対する反応性を検討した。具体的には、CXorf48特異的TCR遺伝子３種を導入したJurkat76.7細胞を用いてそれぞれ前立腺がん細胞株に対するIL-2産生量をELISA法により検討した。PC3細胞株に対して脱メチル化剤添加を行いがん細胞表面上のHLA抗原複合体の発現量を上昇させた上でTCR-T細胞と共培養したところ、2種のTCR-T細胞において少量のIL-2産生が認められた。今後TCR遺伝子の改変を行うことで、より反応性の高いTCR-T細胞の作製を目指していく必要がある。また、上記と並行して免疫細胞染色によるCXorf48タンパク発現検出系の確立を試みた。pMAL発現システムを利用してMBPタグ付きCXorf48タンパクを作製した。さらに精製したタンパクを抗原としてマウスに免疫したところ、マウス抗血清中に抗CXorf48抗体が含まれることをウエスタンブロット法により確認した。CXorf48に対して特に高い反応性を示したマウス抗血清を用いて免疫細胞染色を行った。その結果、前立腺癌細胞株におけるCXorf48の発現が確認された。今後より高い反応性を示す抗体を産生するハイブリドーマを作製しモノクローナル抗体を精製することで、CXorf48を標的としたTCR-T細胞療法が効果を示す患者の探索に応用できる可能性がある。以上の診断治療系の開発により、CRPC患者に対する有効な免疫細胞治療の開発が期待される。 Patients with prostate cancer eventually progress to Castatio Resistat Postate Cace (CRPC). We have previously discovered that CXorf48 antigen gene is highly expressed in prostate cancer stem cells. We have also succeeded in cloning of T cell receptor gene of CXorf48-specific T cells. In this study, we introduced antigen-specific TCR gene into human T cell line and analyzed the response against prostate cancer cells. We transduced 3 kinds of TCR genes into Jurkat 76.7 cells and detected secreted IL-2 by ELISA. 2 out of 3 TCR-T cell lines produced IL-2 against PC3 treated with demethylating agents. Modulation of TCR gene might be needed to improve the efficacy. At the same time, we tried to establish detection method of CXorf48 protein in cancer cells using immunocytochemistry. We purified CXorf48 protein with MBP and immunized mice. Prostate cancer cells were stained with mice serum. Production of monoclonal antibodies using hybridoma will improve the specificity of detection method in order to find patients eligible for treatment targeting CXorf48.The improvement of diagnostic method and immunotherapy against CXorf48 will be useful for CRPC patients.
