• Note (General): type:text; エノール炭酸エステルの脱炭酸反応を起点とする反応活性種の発生に基づく反応開発を行い、本概念を活かした分子間での炭素-炭素結合形成反応を見出した。本申請研究では、以下に示す二つの脱炭酸型分子間炭素―炭素結合形成反応に取り組んだ。 (1)脱炭酸型アリル化反応の開発 申請時、すでにルイス酸を用いるエノール炭酸エステルとアリルシランの反応によって脱炭酸型アリル化が起き、ホモアリルケトン誘導体が生成することを見出していたが、化学量論量のルイス酸を必要としていた。そこで当初、触媒化を目指し、ルイス酸の検討も含めた反応条件の最適化を行った。しかしながら、触媒化は可能であるものの、触媒条件では位置選択性が低下し、結果的に生成物の収率が低下することがわかった。そこで触媒化を断念、基質の一般性を検討し、本反応が様々な基質に適用できることを明らかにした。 (2)脱炭酸型シアノ化反応の開発 申請時、触媒量のルイス酸を用い、エノール炭酸エステルとシリルシアニドの反応によって脱炭酸型シアノ化が起き、シアノケトン誘導体が生成することを見出していた。興味深いことに、上記のアリル化反応とは、同じ基質を用いているにも関わらず、反応の位置選択性が異なる。そこで、反応機構について調べたところ、位置選択性が求核剤の求核力の強さに依存しているという結論を得た。また、中間体として複数のシリルエノールエーテルが生じていることを確認し、塩基性条件での加水分解を反応処理工程に組み込んだところ、収率が飛躍的に向上した。本反応についても様々な基質が適用でき、アリル化反応とは対照的な位置選択性で反応が進行した。 We developed a reaction based on the generation of reaction-active species starting from the decarboxylation reaction of enol carbonate, and found an intramolecular carbon-carbon bond formation reaction utilizing this concept. In this application study, we worked on the following two decarboxylation type intermolecular carbon-carbon bond formation reactions. (1) Development of decarboxylative allylation reaction It was found that decarboxylation-type allylation occurs by the reaction of enol carbonate using Lewis acid and allylsilane to produce a homoallyl ketone derivative, but a stoichiometric amount of Lewis acid was required. Therefore, we initially optimized the reaction conditions including the study of Lewis acid. Although the catalytic reaction is possible, it has been found that regioselectivity is reduced under catalytic conditions, resulting in lower product yields. Therefore, we abandoned catalysis, examined the scope of substrates, and elucidated that this reaction can be applied to various substrates. (2) Development of decarboxylative cyanation reaction It was found that decarboxylative cyanation occurs by the reaction of enol carbonate and silyl cyanide using a catalytic amount of Lewis acid, and a cyanoketone derivative is obtained. Interestingly, the regioselectivity of the reaction is different from the above allylation reaction, even though the same substrate is used. When the reaction mechanism was investigated, it was concluded that the regioselectivity depends on the strength of the nucleophilicity of the nucleophile. Further, it was confirmed that a variety of silyl enol ethers were generated as intermediates and hydrolysis under basic conditions was incorporated, the yield was dramatically improved. Various substrates could be applied to this reaction, and the reaction proceeded with regioselectivity in contrast to the allylation reaction.
