• Note (General): type:text; イノシトール（4, 5）2リン酸（PIP2）は生体膜に存在し、様々なタンパク質と結合することでシグナル伝達や細胞の形態形成などの生理機能を担う。したがって、PIP2結合タンパク質がどのようにして細胞膜中のPIP2を認識するのかを解明することは、生命現象の理解につながる。そこで本研究では、PIP2結合タンパク質と脂質二重膜中のPIP2との相互作用を定量的に調べるため、PIP2含有リポソームとPIP2結合タンパク質の共沈降実験系を確立した。実施例としてPIP2結合タンパク質であるビンキュリンのtailドメイン（Vt）とPLCδ1のPHドメイン（PLCδ1 PH）を調製し、本研究で確立した共沈降法でPIP2含有リポソームとの結合割合を定量的に調べた。 まず、リポソームを構成する脂質中のPIP2の割合を1%で一定にして、タンパク質溶液に添加するリポソーム量を変えてタンパク質の結合率を調べた結果、Vt、PLCδ1 PHともにリポソーム添加量に依存して結合率が増大した。次に、リポソーム添加量を一定にしてリポソーム中のPIP2の割合を2, 4, 6, 8, 10%と変えて結合率を調べたところ、VtとPLCδ1 PH とでPIP2の割合に応じた結合率増大の様子が異なっていた。Vtでは4%以上のPIP2で結合率が大きく増加し、10% PIP2では用いた全てのタンパク質がリポソームと結合した。一方、PLCδ1 PHではPIP2の割合の増大に伴い結合率が増大し、6%以上のPIP2において結合率が50-60%で飽和した。この結果は、PLCδ1 PHの結合率は単純な1:1結合モデルで説明できるのに対し、VtではPIP2割合依存的な急激な結合率上昇からリポソームへの結合に正の協同性があることを示しており、PIP2結合タンパク質の間で結合様式が異なっていることが分かった。特に、Vtにおいて観測された協同的結合は、生体膜中のPIP2の局在や濃度変動に対するビンキュリンの機能に影響を与えている可能性がある。今後、Vtの協同的PIP2認識様式に焦点を当て、その構造基盤を明らかにする。 Phosphatidylinositol 4,5-bisphophate (PIP2) is a component of biological membranes and is responsible for physiological functions such as signal transduction and cell morphogenesis by binding to various PIP2-binding proteins. Therefore, elucidating how each PIP2-binding protein recognizes PIP2 within membranes leads to better understanding of PIP2-related physiological events. In this study, we established experimental system for cosedimentation of PIP2-binding proteins and PIP2-containing liposomes in order to quantitatively examine the interaction between PIP2-binding proteins and PIP2 in membrane. As an example, we prepared a tail domain of vinculin (Vt) and a PH domain of PLCδ1 (PLCδ1 PH), and quantitatively examined their binding ratios to the PIP2-containing liposomes by the cosedimentation method established in this study. First, we examined the protein binding ratios by changing the amount of liposomes added to the protein solution with keeping the percentage of PIP2 in liposomes constant at 1%, and found that the binding ratios of Vt and PLCδ1 PH increased as the amount of liposomes increased. Next, we examined the protein binding ratios by changing the percentage of PIP2 in liposomes to 2, 4, 6, 8, or 10% with the amount of liposomes kept constant. We found that the binding ratios increased in a manner dependent on the percentage of PIP2, however, the PIP2-dependences were different between Vt and PLCδ1 PH. The binding ratio of Vt increased significantly at PIP2 above 4%, and all Vt proteins used bound to the liposomes at 10% PIP2. On the other hand, the binding ratio of PLCδ1 PH linearly increased as the PIP2 percentage increased, and saturated at 50-60% above 6% PIP2. These results indicate that the PIP2-binding mode differs between Vt and PLCδ1 PH: the binding rate of PLCδ1 PH can be explained as a simple 1:1 binding model, whereas Vt indicates a positive cooperativity. In particular, the cooperative binding to PIP2 observed in Vt may influence its physiological function, in response to the change of the location and/or concentration of PIP2 within membranes. Future work will focus on the cooperative PIP2-binding mode in Vt to elucidate its structural basis.
