• Note (General): type:text; MUC1は上皮細胞の表面に存在する2量体の糖タンパク質で、核内に移行することで、ARやNFkB、βカテニン等といった転写因子を介して癌の進展に関与する。予備的検討としてMUC1とmSWI/SNFクロマチンリモデリング複合体は、前立腺癌の核内で共存することを確認している。今回はMUC1とmSWI/SNFで構成される複合体が前立腺癌の進展に及ぼす意義について検討した。神経内分泌前立腺癌細胞株LNCaP-AIにおいて、mSWI/SNFの構成因子であるBRG1・ARID1Aはいずれも高発現していた。MUC1を抑制するとBRG1・ARID1Aのいずれも発現が抑制された。一方、MUC1の過剰発現は、BRG1・ARID1Aの発現亢進を誘導した。MUC1自体は転写因子ではないので、mSWI/SNFの転写を調整するために、他の転写因子を利用する必要がある。Direct binding assayの結果、MUC1は転写因子E2F1と直接結合することが示された。E2F1の抑制は、BRG1・ARID1Aの発現を抑制した。更にMUC1・E2F1・BRG1の抑制はNOTCH1の発現を抑制した。NOTCH経路は去勢抵抗性前立腺癌が神経内分泌前立腺癌に進展する際のkey pathwayである。またMUC1・E2F1・BRG1の抑制はNANOGの発現を抑制した。NANOGはがん幹細胞関連のタンパクである。これらの結果から、去勢抵抗性前立腺癌が神経内分泌前立腺癌に形質転換する際に、MUC1・E2F1の結合体がmSWI/SNFを誘導し、NOTCH経路の活性化やがん幹細胞能獲得に寄与していることが示唆された。 MUC1 is a dimer glycoprotein on the surface of epithelial cells. MUC1 causes cancer progression by translocating into the nucleus and binding to some transcriptional factors such as AR, NFkB, and β-catenin. Preliminary studies showed that MUC1 makes the complex with mSWI / SNF which works as a chromatin remodeling protein in prostate cancer. In this study, we investigated the significance of the complex composed of MUC1 and mSWI / SNF on the progression of prostate cancer. BRG1 and ARID1A, which are constituents of MUC1 mSWI / SNF, were highly expressed in the neuroendocrine prostate cancer cell line LNCaP-AI as well as MUC1. Suppression of MUC1 suppressed the expression of both BRG1 and ARID1A. On the other hand, overexpression of MUC1 induced upregulation of BRG1 and ARID1A. Since MUC1 is not a transcription factor, it is necessary to utilize other transcription factors to regulate the transcription of mSWI / SNF. The Direct binding assay showed that MUC1 binds directly to the transcription factor E2F1. Inhibition of E2F1 decreased the expression of BRG1 and ARID1A. Furthermore, inhibition of MUC1, E2F1, or BRG1 decreased the expression of NOTCH1. The NOTCH pathway is the key pathway for the progress castration-resistant prostate cancer to progress to neuroendocrine prostate cancer. In addition, inhibition of MUC1, E2F1, or BRG1 decreased the expression of NANOG. NANOG is one of the cancer stem cell-related proteins. These results suggested that the MUC1 / E2F1 complex induces mSWI / SNF and it contributes to activation of the NOTCH pathway and acquisition of cancer stemness.
