近年、気管支喘息の病態において2型自然リンパ球（Group 2 innate lymphoid cells : ILC2）を介した自然免疫の関与が注目されている。今回、我々はTLR7アゴニストのR848がIL-33及びILC2を介した好酸球性気道炎症を改善させることを新たに発見した。 R848の作用機序を明らかにするため、TLR7発現細胞をフローサイトメトリーで評価したところ、TLR7はILC2をはじめとしたリンパ球には発現しておらず、マクロファージや樹状細胞に発現が認められた。特に、IL-33刺激によってArg1やFizz1を高発現する2型の間質マクロファージが著明に増加することが判明し、この細胞がTLR7を高発現し、R848刺激によって多量のIL-27を産生することがわかった。 IL-27は以前我々のグループが同定したILC2に対する抑制性サイトカインの一つであり、IL-27受容体ノックアウトマウスを用いた実験によって、R848が間質マクロファージからのIL-27産生を誘導することでIL-33及びILC2による好酸球性炎症を抑制することを証明した。最近、生体における自然免疫細胞はお互いにリンクし、細胞間ネットワークを形成していることが示唆されており、自然免疫細胞ネットワークとして注目されているが、本研究は自然免疫細胞のネットワークを利用し、間質マクロファージを介してILC2による好酸球性気道炎症を抑制するという新しい概念を提唱した。本研究成果はアメリカ胸部学会の学会誌であるAmerican Journal of Respiratory Cell and Molecular Biologyに掲載され、さらに、日本アレルギー学会、アジア太平洋呼吸器学会学術大会(APSR)や各種研究会(Scientific exchange meeting、佐島シンポジウムなど)など様々な国内及び国際的な学会において研究成果を発表し、大きな注目を受けた。 In recent years, the involvement of innate immunity via group 2 innate lymphoid cells (ILC2) in the pathogenesis of asthma has attracted attention. In this study, we found that the TLR7 agonist, R848, ameliorates ILC2-mediated eosinophilic airway inflammation and that R848 does not act directly on ILC2 but on type 2 interstitial macrophages by flow cytometry. Furthermore, we found that interstitial macrophages stimulated by R848 changed their properties and produced large amounts of IL-27, one of the inhibitory cytokines of ILC2 that we previously identified. Finally, we demonstrated that the suppression of ILC2-mediated inflammation by R848 is counteracted in IL-27 receptor knockout mice. These results suggest that innate immune cells in vivo cooperate with each other and form an intercellular network, which may lead to the development of new therapies targeting the entire network of innate immune cells. The research results were published in the American Journal of Respiratory Cell and Molecular Biology and have been presented at domestic and international conferences, including the Japanese Society of Allergy, the annual meeting of the Asian Pacific Society of Respiratory Medicine (APSR), and various research meetings (Academic Exchange Meeting and Sashima Symposium).
