• Note (General): type:text; ヘマトキシリンエオジン染色（以下HE染色）の原理は、色素と組織成分のイオン結合によると考えられている。そのため染色結果はpHの値に大きく左右される。適正に染色されるpHの範囲を特定し、染色性を保持する条件を数値化、更に条件から外れた際のpHの調整方法を検討した。 当院ではカラッチのヘマトキシリン液を用いているが、pH2.9を超えると共染傾向がみられ、pH3.0を超えると明らかに紫の色味が強い染色結果となった。適正範囲はpH2.7から2.9と考えられた。適正範囲以上のpHとなったヘマトキシリン液に酸を加え、pHを下げた場合染色性は改善するのか追加検討を行った。酸化剤であるヨウ素酸ナトリウムを使用したpH調整では、過酸化になってしまい改善は困難であったが、酢酸と塩酸を使用したpH調整では染色性の改善がみられた。特に塩酸は１滴（約50μL）でpHは0.1程度低下し、pH2.9～3.0で染色性の改善がみられたが、本来の適正範囲であるpH2.7~2.9の範囲まで下げると染まらなくなった。 エオジンは酢酸を加え、pH4.1～4.3の範囲で適正に染色された。酢酸を添加しなかった場合pHは7.0～7.3となり染色不良となった。酢酸を過剰に加えたところ、エオジン液は混濁し、染色性が低下したが、アンモニアを加えると染色性は改善された。 検討結果から、染色前のpH測定はHE染色の精度管理に大変有用であると考えられた。ヘマトキシリンのpHが3.0以上、エオジンが4.1未満の場合エオジンが薄いが、ヘマトキシリンが共染し、一見コントラストよく見える。このように染色結果だけでは判定しにくい場合も、pHを測定することによって、数値をもとに適正か否かの判定が可能となった。適正範囲から外れた際の酸、アルカリ試薬添加によるpH調整は染色性の改善に有用である。しかし適正範囲までpHを調整すると染色性が低下する等の問題も発覚し、調整基準の確立については、継続検討としたい。 Hematoxylin-eosin staining is based on ionic binding of dye and tissue components. Therefore, staining results are greatly influenced by the pH value. We identified the pH range for proper staining, quantified the conditions for maintaining staining, and examined methods for adjusting the pH when conditions deviate from the range. We use Carrazzi's hematoxylin solution, which tends to stain excessively when the pH exceeds 2.9, and when the pH exceeds 3.0, the staining results clearly show a strong violet tint, suggesting that 2.7 to 2.9 is the appropriate range. We additionally examined whether the staining property would improve if the pH was lowered by adding acid to the hematoxylin solution that had reached a pH above the appropriate range. Acetic acid and hydrochloric acid improved staining. In particular, hydrochloric acid showed staining improvement from a single drop, but when the pH range was lowered to 2.7-2.9, which is the original appropriate range, staining was no longer possible. Eosin stained properly in the range of pH 4.1-4.3 with the addition of acetic acid. When acetic acid was added in excess, the eosin solution became cloudy and stained poorly. In this case, the addition of ammonia improved the staining. Measurement of pH before staining was considered to be very useful for accuracy control. When the pH of hematoxylin is greater than 3.0 and that of eosin is less than 4.1, the eosin is light, but the hematoxylin is co-stained and appears to be in good contrast. In such cases where it is difficult to determine the staining result alone, measuring pH can help determine whether the values are appropriate. The pH adjustment by adding acid or alkali reagents when the pH is out of the specified range is useful for improving staining, but the establishment of an adjustment standard is desired and will continue to be studied in the future.
