• Note (General): type:text; 中空タンパク質ナノ粒子TIP60を機能性分子カプセルとするため、TIP60への異種分子の内包と、内包したTIP60の外部表面の化学修飾という2段階の機能化について検討した。まず、TIP60に異なるタンパク質分子を内包できる可能性を検討した。大腸菌内でTIP60と内包したいタンパク質を共発現させた結果、様々なタンパク質を内包できることが明らかになった。小角X線散乱分析により、内包分子のTIP60の内部空間占有率がほぼ100%となるケースも確認され、高密度に異種タンパク質を内包できることがわかった。この手法を用いて蛍光タンパク質(tdTomato)のTIP60への内包にも成功し、従来のTIP60と同様に精製することができた。この蛍光タンパク質内包型TIP60の外部表面にあるシステインチオールを対象として、FITC標識した膜透過促進ペプチドを修飾した。修飾条件は事前検討で最適化した、1分子のTIP60に対して2分子のペプチドを修飾する条件とした。その結果、蛍光タンパク質内包型TIP60も同様に修飾することができた。調製したtdTomato内包型TIP60をヒト培養細胞に添加し、共焦点蛍光顕微鏡で観察した結果、tdTomatoを内包したFITC標識TIP60が細胞内に取り込まれていることを確認できた。このとき添加後から数時間経過しても、tdTomatoの蛍光とFITCの蛍光はほぼ共局在したままであったことから、細胞内でTIP60が速やかに分解されてtdTomatoが漏出しているということはないことが示唆された。さらに、次年度の細胞選択的に送達する方法の検討に向けて、TIP60の表面に連結する予定の、標的細胞固有の受容体に結合するpH応答性抗体をmRNAディスプレイ法により試験管内選択することに成功した。現在、この抗体や天然の受容体リガンドを膜透過促進ペプチドを組み合わせることで、細胞選択的なデリバリーを実現可能か検討をおこなっている。 In order to make the hollow protein nanoparticles TIP60 into a functional molecular capsule, we investigated two-step functionalization: inclusion of foreign molecules in TIP60 and chemical modification of the outer surface of TIP60. First, we investigated the possibility that TIP60 could contain different protein molecules. As a result of co-expressing TIP60 and the protein to be encapsulated in E. coli, we found that various proteins can be encapsulated into TIP60. By small-angle X-ray scattering analysis, we confirmed that the internal space occupancy of TIP60 in the inclusion molecule was almost 100%, and found that heterologous proteins could be included in TIP60 at high density. Using this method, we succeeded in encapsulating the fluorescent protein (tdTomato) in TIP60, and purified it in the same manner as the conventional TIP60. The FITC-labeled membrane permeation-promoting peptide was modified for cysteine thiol on the outer surface of this fluorescent protein-encapsulating TIP60. The modification conditions were optimized in advance to modify two molecules of peptide with respect to one molecule of TIP60. As a result, the fluorescent protein-encapsulating TIP60 could be modified in the same manner as the conventional TIP60. We added the prepared tdTomato-encapsulating TIP60 to human cultured cells and observed its localization with a confocal fluorescence microscope. As a result, we confirmed that the FITC-labeled TIP60 containing tdTomato was incorporated into the cells. At this time, the fluorescence of tdTomato and the fluorescence of FITC remained almost co-localized even several hours after the addition, suggesting that TIP60 was not rapidly degraded in the cells and tdTomato was not leaked out. In addition, pH-responsive antibodies that bind to target cell-specific receptors to be linked to the surface of TIP60 were selected in vitro by the mRNA display method for the study of cell-selective delivery methods in the second year of the plan. Currently, we are investigating whether cell-selective delivery can be achieved by combining this antibody or a natural receptor ligand with the membrane permeation-promoting peptide.
