• Note (General): type:text; 近年の組織工学の発展に伴い、血管内皮細胞の三次元培養によって毛細血管を再生することが可能になった。しかし、細胞が三次元ゲルへランダムに入り込み、毛細血管の形態パターンが自発的に形成されるため、血管形状を人為的にコントロールすることができない。任意の形状で様々な三次元組織・臓器を再生しようとする組織工学の研究において血管形成パターンを精緻にコントロールすることは極めて重要である。そこで、本研究では、超短パルスレーザー技術を利用することによって血管形成を自在に誘導する技術を開発することを目的とした。本研究では、血管内皮細胞が毛細血管を形成・伸長する形状を誘導するために、マイクロ流体デバイスの内部に作製した三次元コラーゲンゲルに超短パルスレーザーを照射・走査することで微小間隙を形成した。この時、コラーゲン繊維を切断するnmスケールのマイクロクラックや、レーザーアブレーションによるµmスケールの間隙を形成するためのレーザー照射条件を検討した。その結果、マイクロ流体デバイスにおいて形成したコラーゲンゲルの内部に10～20 µmの微小間隙を形成させることに成功した。さらに、レーザー加工したゲルを用いてヒト血管内皮細胞を培養可能であることを実証した。これらの結果は、超短パルスレーザーによって三次元ゲルに微細構造を加工し、所望の毛細血管パターンを誘導するための重要な足掛かりとなる。レーザー加工において重要となるパラメータも明らかになってきたため、今後の研究でより高精細な加工を実現するための見通しがついた。当該技術を発展させることによって新たな組織形成手法を提案し、組織工学・再生医療の発展に貢献することが期待できる。 Recent advances in tissue engineering allowed to construct capillaries by three-dimensional (3D) culture of vascular endothelial cells. However, since cells randomly migrate into the 3D gel and the morphological pattern of capillaries is spontaneously formed, vascular morphogenesis cannot be artificially controlled. It is critical to precisely control the vascular formation pattern in the study of tissue engineering that attempts to construct various 3D tissues and organs with desired shapes. Therefore, this study aimed to develop a technology to induce vascular formation in a desired pattern using ultrashort pulse laser. In this study, a microchannel was fabricated within a 3D collagen gel formed in a microfluidic device by irradiating and scanning ultrashort pulse laser to induce the vascular formation pattern. Specifically, the laser irradiation conditions for fabricating nm-scale microcracks by disrupting collagen fibers and µm-scale microchannels by laser ablation were investigated. As a result, we succeeded in fabricating microchannels of 10–20 µm within the collagen gel formed in a microfluidic device. Furthermore, we demonstrated that human vascular endothelial cells can be cultured on a collagen gel with a microchannel fabricated by laser irradiation. These results implicate the possibility to induce the desired vascular formation pattern in a 3D gel by fabricating microstructures with ultrashort pulse laser. Since dominant parameters for laser fabrication have been clarified, it is feasible to fabricate microchannels more precisely in a future study. The development of a novel tissue engineering technology is expected based on the present study, which contributes to the advancement of future tissue engineering and regenerative medicine.
