• Note (General): type:text; 神経細胞は興奮電位を生成する。その際に非補償熱の生成を行うが、この熱生成に伴う温度上昇を精密に計測することができれば、神経細胞でのエントロピー生成を見積もることが可能となる。神経細胞のエントロピー生成に関しては、興奮電位のスパイク列から情報論的なエントロピーの見積りから単一神経細胞が担う情報量を求めることができる。この情報量と熱的な見積もりから得られた情報量との関係の対応づけは、生命情報学の本質的な問題である。 我々はこの熱力学的なエントロピー見積もりを神経細胞について行うために、蛍光タンパク質型温度センサーを神経細胞に導入し、合わせて細胞内カルシムイオンの動員（これは神経活動のマーカーとなる）を可視化するプローブとのデュアルイメージングを行い、神経興奮に伴う温度計測が単一神経細胞で可能であるかを検討した。 ラット海馬神経細胞について、上記２種類の蛍光プローブを細胞内に導入し、適切な蛍光シグナルの分離を行うことにより、細胞内温度計測と神経活動を同時可視化する実験系の構築に成功した。また今回の助成で購入した細胞外液温度を計測するための高精度温度計により細胞外温度を計測することもイメージングと同時行った。 その結果、高濃度KCl刺激やナトリウムイオンイオンチャネルを強制的に開かせる薬物の刺激による一過的な細胞内カルシウムイオン濃度上昇に伴って、単一神経細胞レベルでの温度変化を蛍光プローブにより可視化することに成功した。またこの際に、細胞外溶液でも一過的な温度変化を行うこと、この温度変化は細胞内での温度変化とよく似た挙動を示すことを明らかにした。現在神経細胞やHela細胞を用いて、温度蛍光プローブの変化と実際の温度変化との対応関係を調べ、蛍光変化から細胞内温度を見積もる実験を進めており、温度変化から単一細胞での熱力学エントロピー生成を見積ることが可能になると期待できる。 Neurons generate the heat during action potential generation. If we can precisely measure the temperature rise associated with this heat generation, we can estimate the entropy generation in neurons. As for entropy generation in neurons, the amount of information carried by a single neuron can be determined from an informatic meaning of entropy from a spike sequence of excitatory potentials. Clarifying the relationship between this informatic entropy and the entropy obtained from the thermal estimate is an essential problem in bioinformatics. We have performed this thermodynamic entropy estimation for neurons by introducing a fluorescent protein-type temperature sensor into the neuron and combining it with a probe that visualizes the mobilization of intracellular calcium ions (which is a marker of neuronal activity) for dual imaging to determine the temperature. We examined whether measurement is possible in a single neuron. By introducing the above two types of fluorescent probes into rat hippocampal neurons and fluorescent signal isolation, we succeeded in constructing an experimental system for simultaneous visualization of intracellular temperature measurement and neural activity. We also measured the extracellular temperature using a high-precision thermometer for measuring the temperature of extracellular fluid, which was purchased with this grant, simultaneously with imaging. As a result, we succeeded in visualizing the temperature change at the level of a single neuron by fluorescence probes in response to transient increase in intracellular calcium ion concentration induced by high concentration of KCl or by a drug that force sodium ion channels to open. We found that the transient temperature change also occurs in the extracellular solution and that this temperature change behaves similarly to the intracellular temperature change. We are currently conducting experiments using neurons and Hela cells to investigate the correspondence between the change of fluorescence of the temperature probe and the actual temperature change, and to estimate the intracellular temperature from the fluorescence change.
