• Note (General): type:text; 脱細胞化組織は生体組織から細胞を除去することで得られる三次元的な細胞外マトリクスである．脱細胞化組織を作製する手法の一つである高静水圧印加処理法は，薬剤残存のリスクがなく大型の三次元組織や臓器に適用できる可能性がある．しかしながら，超高静水圧を印加すると細胞外マトリクスに損傷を生じて細胞培養における足場構造としての機能を損なう可能性があり，一方で，低レベルの高静水圧を印加した場合には細胞除去が不十分になる問題がある．そこで，本研究では過冷却保存に着目し，過冷却処理と高静水圧処理を複合化することによって，圧力レベルを下げながら細胞除去率を超高静水圧処理と同等とするような脱細胞化処理技術を確立することを目的とした．本年度は細胞懸濁液および三次元培養体を対象として過冷却保存後に高静水圧を印加することによる影響について検討した． まずはじめに，細胞懸濁液を凍結保護剤フリー下で過冷却保存することによる影響について検討した．過冷却デバイスを用いて細胞懸濁液を凍結状態，冷蔵状態，および過冷却状態で0〜36時間保存して，細胞生存性，初期接着性および細胞増殖性に与える影響を明らかにした． 次に過冷却保存と静水圧印加の複合化が細胞構造および細胞機能に与える影響について調査した．過冷却処理チャンバ内で細胞懸濁液を過冷却保存後100, 150, 200 MPaの高静水圧を印加し，細胞膜の微細構造，細胞生存性，初期接着性，および細胞増殖性について過冷却保存による前処理の有効性を実証した． 最後に，過冷却保存と高静水圧の複合処理が細胞外基質の微細構造を変性しないことを実証するために，細胞含有コラーゲンゲルに対する過冷却保存と静水圧の複合化が与える影響について調査した．細胞含有コラーゲンゲルを作製し，過冷却チャンバ内で過冷却保存後100, 150, 200 MPaの静水圧を印加した後に，細胞生存性，ゲル内部のDNA残存率，およびコラーゲン繊維の構造について非保存群および冷蔵保存群と比較することで，本研究で提案する脱細胞化処理技術の有効性を実証した． Decellularized tissue is a three-dimensional extracellular matrix obtained by removing cells from biological tissues. High hydrostatic pressure (HHP) treatment, one of promising methods for generating decellularized tissues, has the potential to generate a large three-dimensional tissues or organs without the risk of reagent residues. However, the application of ultra-HHP treatment might damage the extracellular matrix to lose its function as a cell-culture scaffolds, while the application of a low level HHP might result in non-sufficient cell removal. Therefore, in this study, we focused on supercooling preservation and aimed to establish a decellularization treatment technology by the HHP treatment combined with supercooling preservation. In this year, we examined the effects of applying HHP to cell suspensions and 3D cultures after supercooling pre-treatment. Firstly, the effects of supercooling on cell suspensions under cryoprotectant-free conditions were examined. Cell suspensions were stored in frozen, chilled, and supercooled conditions for 0-36 hours using a supercooling device to determine the effects on cell viability, initial adhesion, and cell proliferation. Next, the combined effects of HHP application and supercooling on cell structures and functions were investigated. The cell suspensions were stored under the supercooled condition and then subjected to HHP of 100, 150, and 200 MPa to demonstrate the effectiveness of pretreatment with supercooling on cell membrane, viability, proliferation, and initial adhesion. Finally, to demonstrate that combined treatment with HHP and supercooling does not denature the microstructure of extracellular matrix, the effects of combined treatment on cell-containing collagen gels were investigated. The cell-containing collagen gels were prepared and subjected to the supercooling treatment followed by HHP application at 100, 150, and 200 MPa. From the results, the effectiveness of this method was demonstrated.
