• Note (General): type:text; 思春期特発性側弯症 (AIS)のGenome-wide association study (GWAS)により得られた、Chr.7p22.3の領域についてin vitro解析を行なった。この領域ではlinkage disequilibrium ＞0.8の28個のAISに関連すると考えられるSNPを確認した。まずこれをin silico解析により、AISの発症と有意に関連するゲノム領域内のバリアント３つに絞り込んだ。その３つのバリアントが影響与えると考えられる同領域の遺伝子の発現への影響をCRISPR-dCas9-KRABの系にて調べた。これによりrs78148157周辺配列がUNCXの発現に影響を与えることが分かった。同バリアントに対しEMSAにて、アレル間の結合の強さの差の解析、およびアレルに結合する転写因子の同定を行った。この解析により、rs7814815のAISリスクバリアントを含む配列には、EGR1という転写因子が結合することが分かった。このAISリスクバリアント周辺配列がUNCX遺伝子のプロモーター活性へ与える影響を神経系細胞であるA172を使用したレポーターアッセイにて解析し、AISリスクバリアントはUNCX遺伝子の発現を上昇させることが判明した。(p=0.027) 更に、EGR1をこの系で共発現させると、UNCX遺伝子の発現量の差はリスクアレルとノンリスクアレルで更に増強した。このことから、AISのGWAS関連ゲノム領域のバリアントが、転写因子EGR1を介してUNCXの発現を調整していることが明らかとなった。 In vitro実験にて、AISのリスクバリアントではUNCX遺伝子の発現量が上昇していることが期待されたため、In vivoの実験を計画した。ゼブラフィッシュ胚に対してUNCXの発現量が上昇するように mRNAのマイクロインジェクションを行なった。するとUNCX mRNAを投与されたゼブラフィッシュでは、その投与量依存性に脊椎の奇形、体軸の曲がりを確認した。UNCXの発現上昇はゼブラフィッシュで体軸形成不全を生じた。 In vitro analysis was performed on the region of Chr.7p22.3 obtained by the Genome-wide association study (GWAS) of idiopathic adolescent scoliosis (AIS). In this region, 28 SNPs with linkage disequilibrium> 0.8 were candidates SNPs thought to be related to AIS pathogenicity. First, in silico analysis narrowed down the candidate variants into three variants in the genomic region. The effects of the three variants on the expression of genes in the region were investigated using the CRISPR-dCas9-KRAB system. This analysis revealed sequence around rs78148157 affects the UNCX expression. For the rs78148157, EMSA was used to analyze the difference in binding strength between alleles and identified transcription factors that bind to alleles. In this assay, a transcription factor EGR1 bind to the AIS risk variant of rs7814815. The effect of the AIS risk variant on the promoter activity of the UNCX gene was analyzed by a reporter assay using A172, which is a nervous tissue cell. The AIS risk variant increased the expression of the UNCX gene. (p = 0.027) Furthermore, when EGR1 was co-expressed in this system, the difference in the promoter activity of the UNCX gene was further enhanced between the risk allele and the non-risk allele. This revealed that a variant of the GWAS-related genomic region of AIS regulates UNCX expression via the transcription factor EGR1. In vivo experiment was performed using zebrafish embryo since the expression level of the UNCX thought be increased in the AIS patient. Microinjection of UNCX mRNA was performed on zebrafish embryos so that the expression level of UNCX was increased. In the zebrafish with UNCX mRNA was administered, spinal malformation and body axis bending were confirmed in dose dependent manner. The increased expression of UNCX resulted body axis deformation in zebrafish.
