• Note (General): type:text; 昆虫比較発生学において、卵の定常的な確保のための昆虫の安定的な飼育・繁殖法と、卵の固定・観察法の確立は必要不可欠であり、化学的手法の改良や検討は最重要課題である。本課題では、ゴキブリ類の比較発生学的研究のための化学的基盤構築を目指し研究を行なった。 本年度も引き続き、野外棲息種で、発生学的知見が皆無であるチビゴキブリ科のクロモンチビゴキブリAnaplecta japonicaを対象に、基本データとなる本種の生物学的知見の蓄積および卵の固定法を初めとする化学的手法の検討を行った。 愛媛県での採集により得られた成虫およそ130個体を飼育し、飼育系より得られた卵鞘から最適な固定法の探索と検討を行った。その結果、Karnovsky固定液やパラフォルムアルデヒドなどのアルデヒド系の固定液が組織観察において良好な結果をもたらすことが分かった。また、卵鞘の解剖技術の一つとして、酵素溶液による卵鞘殻の軟化法を取り入れた。これにより、卵へのダメージを抑えつつ、卵と卵鞘との接着を容易に離すことができるようになった。1卵鞘あたりには4-6卵が含まれ、卵鞘内で卵腹面を向かい合わせるように配置している。卵は長さ1.2 mm, 幅0.6 mmで黄白色を呈する。得られた卵を、ナノスーツ法走査型電子顕微鏡観察および卵殻のみのプレパラート観察を行った結果、卵腹面後方に30-40個の漏斗型の卵門（精子の侵入孔）が局在することが明らかとなった。また、産卵から孵化までの各発生段階の卵を固定し、光学顕微鏡観察ならびに組織切片の観察によって、胚発生の概略を把握するとともに、胚運動型が、胚発生の過程で胚の頭尾軸の逆転を伴う「胚軸逆転型」であることが明らかとなった。本研究により、ゴキブリ類の中でもクロモンチビゴキブリを対象として、発生学的観察により有効な卵の固定・観察法が確認された。今後は本種の胚発生データの補完を行うとともに、これまでの研究成果の公表を目指す。 n insect comparative embryology, it is important to establish chemical methods for gaining insect eggs steadily and maintaining insect culture. Thus, this project aims at constructing and improving the methods for comparative embryology of Blattodea (cockroaches). As a material of the study, the blattodean group Anaplectidae, for which embryological knowledge was totally lacking, was examined using Anaplecta japonica Asahina, 1977. 130 adults of A. japonica collected from Ehime prefecture of Japan were reared, and their oothecae and eggs were chemically examined. In the fixation for eggs of A. japonica, Karnovsky's fixative or 4% paraformaldehyde brought about fine resolution in histological observations. As for the dissection of oothecae (egg cases), employing enzyme solution containing protease and lipase, the oothecal shells were softened so that eggs were easily taken from them. The ootheca contains 4-6 eggs, which are arranged in two rows with their ventral side facing each other. The eggs are ellipsoidal, ca. 1.2 mm and 0.4 mm wide, and whitish yellow. Fixed eggs were processed with the nano-suit method and observed under high-vacuum SEM, and the chorions torn out from eggs were mounted on the slide glasses and observed under a biological microscope. 30-40 micropyles, which are small pores for the entrance of sperm, are grouped in the posterior region of the ventral side of the eggs. For the examination of embryonic development, the fixed eggs of each developmental stage were histologically processed and observed under a biological microscope. The present study revealed that the embryos of A. japonica undergo the development with their anteroposterior axis reversed. That is the blastokinesis in Anaplectidae is of the "reversion type". As has been addressed, this study found an effective chemical method for fixation of cockroach eggs, especially in A. japonica. Using this method, further embryological examination would be conducted for publishing those results.
