• Note (General): type:text; 酸化脂肪酸は、ヒドロキシ基やエポキシ基等の酸化修飾基を有する脂肪酸であり、酸化脂肪酸を含むリン脂質分子種は「酸化リン脂質」と総称される。生体内には多種多様な酸化リン脂質分子種が存在し、イオンチャネルなどの膜タンパク質の機能を制御することが知られている。一方で、これら酸化リン脂質の生合成経路や組織内分布については、不明な点が多い。そこで本研究では、酸化リン脂質の生合成を担う酵素の同定と、イメージングMSを用いた酸化リン脂質の組織内分布の可視化の手法開発を試みた。 まず、飽和脂肪酸や不飽和脂肪酸をリン脂質にアシル化する酵素であるアシル基転移酵素に注目した。特にアラキドン酸などの高度不飽和脂肪酸を好んで基質にする既知のアシル基転移酵素を4分子について、コムギ無細胞タンパク質合成系を用いて、リポソーム上に合成することに成功した。合成したアシル基転移酵素のプロテオリポソームは、いずれもアラキドン酸をリン脂質に導入する活性を有しており、酵素として機能することを確認した。次に、これら4種類のアシル基転移酵素のプロテオリポソームが実際に酸化脂肪酸をリン脂質に導入する活性を有するかを調べたところ、4種類のうち1種類のアシル基転移酵素が、ヒドロキシ酸化脂肪酸をホスファチジルコリンにアシル化する活性を有することを見出した。この活性は、酵素活性中心と思われるヒスチジンをアラニンに置換することで、完全に消失した。以上より、本研究によりヒドロキシ酸化リン脂質の合成を担うアシル基転移酵素の同定に成功した。 次に、当該ヒドロキシ酸化リン脂質の組織中の空間分布をイメージングMSで解析する系の構築を試みた。このヒドロキシ酸化リン脂質は標準品が市販されていない。そこで、当該ヒドロキシ酸化リン脂質を大量に含む細胞を樹立する必要があり、上述のヒドロキシ酸化リン脂質の合成を担うアシル基転移酵素の過剰発現細胞株を樹立した。 Oxidized phospholipids contain oxidized fatty acids such as hydroxyl or epoxy fatty acids. Our body and cells possess a variety of oxidized phospholipid species. The functions of membrane proteins like ion channels are controlled by the oxidized phospholipids through the dynamic changes of their surrounding membrane environments. Although the patho-physiological significances of the lipids are well recognized, their synthetic pathways and tissue distributions remain unclear. In this study, we aimed to identify the responsible genes for the oxidized phospholipids synthesis and to establish the methods for imaging mass spectrometry (MS) for the lipids. We first reconstituted known lysophosholipids acyltransferases (LPLATs) on liposomes by making use of wheat germ extract. The LPLATs exhibited the activities for introduction of arachidonic acids into lysophosphatydilcholine as reported previously, indicating the quality of the LPLAT proteoliposomes. Among the four different LPLATs, we found that a LPLAT possesses the enzymatic activities for the acylation of hydroxyl arachidonic acid into phosphatidylcholine. To establish the imaging MS system for the oxidized phospholipid, we successfully established the cell lines which overexpress the LPLAT. We are planning to use the lipids extracted from the LPLAT-expressing cells for imaging MS because the the oxidized phospholipid is not commercially available.
