• Note (General): type:text; 刺激因子の2型サイトカイン（IL-5, IL-4, IL-33）、1型サイトカイン（TNF-alpha, IFN-gamma）の存在下で培養した健常者の血中好酸球を用いてRNA-seqを実施した．RNA-seqにより発現上昇因子として、IL-5とIL-4に共通するGGT5、IL3RA、IL-5特異的なIL2RA、CCL23、IL-4特異的なTGM2、IL1RL1、IL-33とTNFに共通するC3、CCL4、IL1、IFN特異的なGBP、TRIMなどを同定した．同様の変化をプロテオミクスとフローサイトメトリーで確認し、これらの因子が蛋白翻訳後に機能的分子となりうることを示した．リピドミクスではIL-4とIL-5の刺激によりLTD4の産生量が相乗的に増加することを確認し、発現したGGT5などの脂肪酸代謝酵素が実際に脂肪酸代謝パターンを変化させ、特定の脂質メディエーターの産生に寄与することが証明された． 私たちが保有している鼻茸好酸球の遺伝子発現データを用いて、これらの刺激因子による変化が実際の炎症部位の好酸球で生じているかどうか検証した．IL-5, IL-4, IL-33, TNF-alphaによる変化と同じ遺伝子発現パターンの変化が複合的に生じていることが確認された．IFN-gammaの刺激によって変化する因子については発現量の増減が均一ではなかった．以上から、IL-5, IL-4, IL-33, TNF-alphaが病態に関与している可能性が示唆された．IFNについては病態への寄与度は不明であり、今後他のIFN-alpha/beta, IFN-lambdaを用いた比較検討が必要であると考えられる． 喘息の喀痰細胞の遺伝子発現パターンを示したデポジットデータをGEOから入手し、IL-4・IL-5とこれらのサイトカイン刺激により好酸球で誘導された遺伝子との相関関係を調べた．両サイトカインはGGT5, IL3RA, IL2RA, CCL23, TGM2, IL1RL1と相関を認め、特にIL-4との相関係数が高い傾向を認めた．本研究で同定された因子群の変化は副鼻腔炎、喘息などの様々なアレルギー疾患の病態の解釈に有用である可能性があり、今後も解析を進める予定である． RNA-seq was performed using blood eosinophils from healthy subjects cultured in the presence of eosinophil stimulatory factors, type 2 cytokines (IL-5, IL-4, IL-33) and type 1 cytokines (TNF-alpha, IFN-gamma). This analysis revealed the upregulated factors including IL-5 and IL-4-induced GGT5 and IL3RA, IL-5-induced IL2RA and CCL23, IL-4-induced TGM2 and IL1RL1, IL-33 and TNF-induced C3, CCL4, and IL1, and IFN-induced GBP and TRIM. Similar changes were confirmed by proteomics and flow cytometry, indicating that these factors can become functional molecules after protein translations plus upregulation of gene expressions. In lipidomics, we confirmed that stimulation of IL-4 and IL-5 synergistically increased LTD4 production, proving that expression of fatty acid metabolism-related enzymes such as GGT5 actually altered fatty acid metabolism characterized by selective production of bioactive lipid mediators. Using our gene expression data of nasal polyp-derived eosinophils, we verified whether the changes induced by these stimulators occurred in eosinophils at the actual site of inflammation. This analysis confirmed that the changes in gene expression patterns were complexed with those induced by IL-5, IL-4, IL-33, and TNF-alpha. The gene expression patterns altered by IFN-gamma were not uniformly changed. The contribution of IFN to the pathogenesis of nasal polyp is unknown and comparative studies using other IFNs such as IFN-alpha/beta and IFN-lambda are needed. Deposit data showing gene expression patterns of sputum cells taken from patients with asthma were obtained from GEO. The correlation between IL-4 or IL-5 and the genes in eosinophils induced by stimulation with these cytokines was examined. Gene expressions of both cytokines were correlated with GGT5, IL3RA, IL2RA, CCL23, TGM2, and IL1RL1. The correlation with IL-4 was higher than that with IL-5. The changes in the factors identified in this study may be useful in interpreting the pathogenesis of various allergic diseases such as sinusitis and asthma and we plan to continue our analysis in the future.
