• Note (General): type:text; TIP60は60分子のタンパク質が自発的に会合して中空サッカーボール形状に組み上がった分子であり、そのボールの表面には2.4 nm直径の孔が20箇所もあり、小分子化合物は素通りできるような構造を有している。しかし、表面の孔よりも大きい中分子の内包は実現できていなかった。この実現には一度60分子をバラバラに分解し、その後、再びサッカーボール状に会合させる仕組みが必要であった。そこで、60分子が会合できなくなるような変異体の探索を行った。その結果、会合できなくなるK67E変異体が発見された。さらに、この変異体にバリウムイオンを添加すると、元の60量体が得られることが明らかになった。また、同時にDNAなどを添加しておくことで、内部空間に閉じ込められることも明らかにした。 こうした分子材料は、一般に調製コストが高くなる傾向があり、結果的に実用化を阻む壁となっている。特にタンパク質の精製では、時間がかかり、かつスケールアップも行いにくいカラムクロマトグラフィーが必要なケースが多く、TIP60も例外ではなかった。そこで、通常の精製方法とは異なる大量精製法の開発に着手し、カラムクロマトグラフィーを利用することなく、従来の10倍以上のTIP60を一段階で獲得できる手法も確立した。 一方、独自の膜透過促進ペプチドを用いて、ペプチドや核酸からなる中分子医薬を細胞内にデリバリーするための予備的検討をおこなった。まず、膜透過促進ペプチドと細胞表面受容体に結合する抗体を組み合わせることで、ペプチド医薬を受容体発現細胞選択的に細胞質送達することに成功した。また、核酸医薬として期待されているsiRNAをAgo2との複合体として、膜透過促進ペプチドを用いて細胞質に送達することで、従来法よりも迅速なノックダウンが可能となることを見出した。今後、膜透過促進ペプチド、抗体とTIP60を組み合わせることで、ペプチドや核酸を細胞選択的にデリバリーすることが期待できる。 TIP60 is a molecule in which 60 molecules of protein spontaneously associate to form a hollow soccer ball, and the surface of the ball has 20 pores with a diameter of 2.4 nm through which small molecules can pass through. However, encapsulation of middle molecules larger than the pores on the surface has not been realized. To realize this, it was necessary to have a mechanism that once dissociated the 60 molecules and then reassembled them into a soccer ball shape. Therefore, we searched for mutants that could not associate with 60 molecules. As a result, a K67E mutant was discovered that was unable to associate. Furthermore, it was found that addition of barium ions to this mutant yielded the original 60-mer. Furthermore, we demonstrated the encapsulation of single-stranded DNA molecules in the internal space. Such molecular materials generally tend to be expensive to prepare, which is a barrier to their practical application. Protein purification in particular often requires column chromatography, which is time-consuming and difficult to scale up, and TIP60 was no exception. Therefore, we started developing a large-scale purification method that is different from the usual purification method, and established a method that can obtain more than 10 times more TIP60 in a single step without using column chromatography. On the other hand, we conducted preliminary studies for intracellular delivery of middle-molecule drugs composed of peptides or nucleic acids using our membrane-permeation-enhancing peptides. First, by combining a membrane permeation-enhancing peptide and an antibody that binds to a cell surface receptor, we succeeded in selectively delivering a peptide drug to the cytoplasm of receptor-expressing cells. In addition, we found that siRNA, which is expected as a nucleic acid drug, can be knocked down more quickly than conventional methods by complexing siRNA with Ago2 and delivering it to the cytoplasm using a membrane-permeation-enhancing peptide. In the future, it is expected that cell-selective delivery of peptides and nucleic acids will be possible by combining membrane-permeation-enhancing peptides, antibodies, and TIP60.
