• Note (General): type:text; 我々は免疫原性細胞死(ICD)に着目し、がん細胞に作用して患者の抗がん免疫反応を増強し得る天然由来化合物の同定を試みた。42種の新規天然化合物群を用いて、MUM24細胞をモデルケースとしてICDを誘導する化合物の探索を行った。まず、WST-1 assayにより、5μMおよび50μMで細胞増殖抑制能を有する化合物を6種類抽出した。次に薬剤処理後のがん細胞においてICDマーカーであるCalreticulin(CRT)表出をflow cytometryにより検討したところ、低濃度でCRT発現を誘導し得る２種の化合物X、Yを抽出した。続いて、Western blot法にてもう一つのICDマーカーであるHigh Mobility Group Box 1(HMGB1)の細胞外放出を検討したところ、がん細胞からのHMGB1の放出量増加が認められた。この際化合物Ｘについては類縁体2種についてもHMGB1の細胞外放出を検討したが、リード化合物Ｘと有意差は認められなかった。さらに化合物Xで処理したMUM24細胞をヒト末梢血単核球由来樹状細胞（DCs）と共培養すると、活性化マーカーのCD80とHLA-DRが顕著に増加し、DCsの成熟が観察された。以上より、化合物X、YはICD誘導能を有すると考えられた。化合物XはICD誘導因子である活性酸素の産生を促進することが報告されているため、抗酸化剤を化合物Xに併用したところ、CRTの表出は抑制されなかった。従って、化合物XによるICDは活性酸素を介さない新たな機序で起こっていることが推測された。さらに、化合物X添加前後のMUM24よりtotal RNAを抽出し、RNAシークエンスによりICDを誘導する遺伝子変化について解析中である。化合物Ｙについては、Panc1細胞に添加したところ、5μMおよび50μMにおいてCRT表出が増強した。今後、これらの化合物の免疫修飾作用について作用機序の解明を行うと同時に、正常細胞への毒性の検討など、膵がん細胞に対する臨床応用を目指した基礎データを積み重ねていく。 We have focused on immunogenic cell death (ICD), and tried to identify natural products which could enhance anti-cancer immune response of patients. First, we checked cytotoxicity of 42 novel natural products against MUM24 cells and found 6 candidates. We next evaluated cell surface expression of calreticulin (CRT) after treatment with these products. Compound X and Y were found to induce CRT with low dose. We could also detect secretion of High Mobility Group Box 1(HMGB1) from MUM24 treated with those compounds. Then we evaluated maturation of dendritic cells (DCs) co-cultured with compound-treated cancer cells and found that expression of CD80 and HLA-DR increased. These results suggest that X and Y are ICD Inducers. Compound X has been reported to produce ROS, however, we found that CRT expression was not abolished by addition of anti-oxidants, suggesting that there are other new pathways of ICD induced by the natural products. Therefore, we are now analyzing gene expression of MUM24 after treatment with compound X. We also found that CRT expression increased in PANC1 cells after treatment with compound Y. We are going to study the precise mechanism of immunomodulatory effects of these compounds. At the same time we will evaluate the effects of these compounds on normal cells and obtain basic data for the clinical use of these compounds in pancreatic cancer.
