• Note (General): type:text; 本研究おいて、慶應義塾大学・医学部・病院リウマチ・膠原病内科に所属する研究分担者のこれまでの研究により得られた肉芽腫性炎症部位に高発現する候補分子Xを標的として、この候補分子Xの発現を検出可能な新たな蛍光プローブの開発に取り組んだ。候補分子Xは蛋白質の切断活性を有する酵素であることから、まずは候補分子Xにより選択的に切断される基質認識部位のアミノ酸配列を調べることとした。研究分担者らとのディスカッションを通して、これまでに報告されている論文やデータベースを活用して、候補分子Xの天然基質を含めた選択的な基質アミノ酸配列の候補配列を23個選定した。さらに、ペプチド合成機を研究室内に新たに立ち上げ、これらの候補アミノ酸配列を、本装置を用いて、Rink amide-MBHA Resin上でFmoc固相合成法によって合成することに取り組んだ。また、全ての合成したペプチドに関して、アミノ酸配列のN末端に蛍光色素を結合させて、蛍光標識した。この蛍光標識は、これから行う酵素アッセイにおいて、酵素反応の進行の評価に際してHPLCを用いるため、ペプチド分子を蛍光標識することで高感度に酵素反応の進行を評価することができる。固相から切り出した蛍光標識ペプチドは、エーテル沈殿によって粗精製した後、HPLCにて精製を行った。また、MALDI-MSで合成したそれぞれのペプチドの分子量を測定することでペプチドの構造の確認を行った。また、コントロール配列として、候補分子Xによる切断活性は高いが選択性の低い2個の既知のペプチドも、ペプチド合成機を用いて合成を行った。このようにこれまでに、候補ペプチド23個とコントロールペプチド2個の計25個の蛍光標識ペプチドを合成し、全て粗精製まで完了している。そのうち、5個はHPLCによる精製も完了した。今後は、これまでに合成した蛍光標識した全てのペプチドを候補分子Xとin vitroで酵素反応させ、HPLCによって切断効率を評価する。また、それらの酵素選択性も評価する予定である。 The co-investigators, who belong to division of rheumatology department of internal medicine Keio university school of medicine, have found the candidate molecule X highly expressing in the granulomatous inflammatory site for its diagnosis so far. In this study, we try to develop a novel fluorescent probe for detecting the expression of the candidate molecule X. The candidate molecule X is an enzyme which cleavages the specific amino acid sequences, so we firstly searched the amino acid sequences which are selectively cleaved by the candidate molecule X. Through the discussion with the co-investigators, we determined 23 candidate amino acid sequences as candidate molecule X's selective substrate sequences including the endogenous substrates on the basis of the previous reports and data bases. Further, we newly set up the peptide synthesizer in our laboratory, and tried to synthesize these candidate peptides with Rink amide-MBHA Resin by using Fmoc solid-phase peptide synthesis. We also conjugated the fluorescent dye to the N-terminal of each synthesized peptide. This fluorescent labelling was expected to be useful to high-sensitively monitor the enzymatic reaction with the candidate molecule X, because this enzymatic assay is performed by HPLC analysis. So far, we roughly purified each synthesized peptide cleaved from the solid phase by the precipitation in ether, then further purified some of these peptides by HPLC. The structures of the purified peptides were verified by MALDI-MS analysis. Furthermore, we also synthesized 2 control peptides which are high-efficiently but low-selectively cleaved by the candidate molecule X by using the peptide synthesizer. Thus, we succeeded in the synthesis and rough purification of total 25 fluorescently labelled peptides including 23 candidate peptides and 2 control peptides so far. 5 candidate peptides among them have been already highly purified by HPLC. In the next year, we will perform the enzymatic assay of all the fluorescently labelled synthesized peptides with the candidate molecule X in vitro, and will assess the efficiency of the enzymatic reaction with them by HPLC. We will also assess the enzymatic selectivity of all the synthesized peptides.
