• Note (General): type:text; 尿路上皮癌や多発性骨髄腫でFGFR3（線維芽細胞増殖因子受容体3）が過剰発現していることが報告されている。FGFR3阻害抗体には一部、抗腫瘍効果があることが報告されているが、より強力な次世代型の抗体医薬品（二重特異性抗体やADC、α線核種標識抗体）としての開発は行われていない。我々はFGFR3に対する次世代型の抗体医薬品を開発するために、FGFR3に対する複数のモノクローナル抗体を作製した。先ず、FGFR1、FGFR2、FGFR3b、FGFR3c、FGFR4のFc融合タンパク質を精製した。Balb/cマウスにFGFR3c-Fcタンパクを免疫し、免疫後、慢性骨髄性白血病細胞株であるK562細胞と反応する抗血清が含まれることを確認した。抗血清が含まれるマウスから脾臓を摘出し、Sp2/O-Ag14細胞と細胞融合装置ECFG21を用いて電気融合を行なった。FGFR3cあるいは3bの両方に特異的に反応し、FGFR1、2、4に反応しないハイブリドーマをELISA法により評価した。その後、K562細胞および中皮腫細胞株であるMESO4細胞を用いたFACSによる解析において、K562細胞への強い結合が確認された、IgG1由来の1H10と5D5、IgM由来のA1F7についてセルクローニングを行い、5'-RACEにより一次配列情報を取得した。その配列情報を用いて、ヒトキメラ抗体を発現するベクターを構築し、医薬品生産用CHO細胞により大量精製した。ヒトキメラ抗体のFGFR3への親和性をK562細胞を用いたFACSにより評価した結果、いずれのヒトキメラ抗体もマウスモノクローナル抗体より高い親和性が確認された。次に、BIACOREによる解離定数を測定した結果、1H10が4.67x10^-9、5D5が1.97*10^-8であることが判明した。以上より、次世代型の抗体医薬品に適応可能なFGFR3抗体の作製に成功した。 Overexpression of FGFR3 (fibroblast growth factor receptor 3) has been reported in urothelial carcinoma and multiple myeloma. Although FGFR3 inhibitory antibodies have been reported to have some anti-tumor effects, development as more potent next-generation antibody drugs (bispecific antibodies, ADCs, alpha-ray radionuclide-labeled antibodies) has not been conducted. To develop next-generation antibody drugs against FGFR3, we generated several monoclonal antibodies against FGFR3. First, we purified Fc fusion proteins of FGFR1, FGFR2, FGFR3b, FGFR3c, and FGFR4. Balb/c mice were immunized with FGFR3c-Fc protein and, after immunization, were confirmed that they contained antiserum that reacted with K562 cells, a chronic myeloid leukemia cell line that has already been reported to express FGFR3. Spleens from mice containing the antiserum were removed and electrofused with Sp2/O-Ag14 cells using the ECFG21 cell fusion device. Hybridomas that reacted specifically to both FGFR3c or 3b but not to FGFR1, 2, or 4 were evaluated using ELISA. Subsequently, cell cloning was performed on IgG1-derived 1H10 and 5D5 and IgM-derived A1F7, which were found to bind strongly to K562 cells but not to mesothelioma cell line MESO4 cells by FACS analysis, and primary sequence information was obtained by 5'-RACE. Using the sequence information, a vector expressing human chimeric antibody was constructed and mass purified by CHO cells for drug production. The affinity of the human chimeric antibodies to FGFR3 was evaluated by FACS using K562 cells, and both human chimeric antibodies showed higher affinity than the mouse monoclonal antibody. Next, the dissociation constants by BIACORE were determined to be 4.67x10^-9 for 1H10 and 1.97*10^-8 for 5D5. Based on the above, we have succeeded in producing an FGFR3 antibody that can be adapted to next-generation antibody drugs.
