• Note (General): type:text; 後発白内障は白内障手術後に再び視機能が低下する疾患であり、手術後に残存した水晶体上皮細胞が間葉系細胞へと形質転換 (EMT) し、水晶体嚢が濁ることで発症する。これまで本講座では、Capsaicinが水晶体上皮細胞のEMTを抑制することを明らかにしてきた。しかしその詳細な分子メカニズムは解明されていない。そこで本研究では、ヒト水晶体上皮細胞株 (ihLEC-NY2) を用いてCapsaicinのEMT抑制メカニズムをCalpains-PTPs相互作用に着目し検討した。 ihLEC-NY2にTGFb2で刺激しEMTを誘導した。RT-PCRにより、CalpainsやPTPsの発現を検討した。発現が認められたCalpainsを過剰発現させたihLEC-NY2を用い、CapsaicinのEMT抑制効果を検討した。その結果、ihLEC-NY2において、Capsaicinは、TGFb2誘導Smad2/3, ERK1/2, EGFRのリン酸化を抑制した。この抑制はTRPV1チャネル阻害剤で解除されたことから、TRPV1チャネルの活性化を介してEMTを抑制することが推察された。RT-PCR法より、水晶体上皮細胞にはCalpain1, Calpain2, CalpainS1, PTPN1, PTPN2, PTPN9が発現していることが認められた。Calpain2, CalpainS1を過剰発現させた細胞では、CapsaicinによるTGFb2誘導EMTがcontrolに比べ強く抑制されたことからCalpain2, CalpainS1の関与が推察された。さらにCalpainsの切断サイトを同定するため、Peptidyl-MCAを用いて、ペプチド基質の分解活性を評価した。その結果、Capsaicin添加によりAla-Ala-Phe配列が強く切断されたことから、PTP1Bが18, 384番目で切断される可能性が示唆された。 以上の結果より、TRPV1チャネルの活性化を介したEMT抑制にCalpain2, CalpainS1が関与することが推察された。今後は、カプサイシンをリード化合物としてより刺激性の少ない後発白内障抑制薬の創製を目指す。 Secondary cataract is a condition in which visual function deteriorates again after cataract surgery, and occurs when the lens epithelial cells remaining after surgery transform into mesenchymal cells (Epithelial-Mesenchymal Transition: EMT), resulting in clouding of the lens capsule. We have previously shown that capsaicin suppresses EMT of lens epithelial cells. However, the detailed molecular mechanism has not been elucidated. In this study, we investigated the mechanism of EMT inhibition by capsaicin using a human lens epithelial cell line (ihLEC-NY), focusing on the Calpain-PTPs interaction. The expression of Calpains and PTPs was examined by RT-PCR. The EMT suppression effect of capsaicin was examined using ihLEC-NY overexpressing Calpains, which were found to be expressed in human lens epithelial cells. We found that capsaicin suppressed TGFb2 induced phosphorylation of Smad2/3, ERK1/2, and EGFR in ihLEC-NY. This inhibition was lifted by TRPV1 inhibitor, suggesting that capsaicin suppresses EMT via activation of TRPV1 channel. In the cells overexpressing Calpain2 and Calpain S1, TGFb2-induced EMT was more strongly suppressed than un the control cells, suggesting Calpain2 and CalpainS1 could be important proteins to suppress the EMT by capsaicin. To further identify the cleavage site of calpains, we evaluated the degradation activity of peptide substrates using peptidyl-MCA. The results showed that the Ala-Ala-Phe sequence was strongly cleaved by the addition of capsaicin, suggesting that PTP1B bay be cleaved at sites 18 and 384. These results suggested that Calpain2 and CalpainS1 are involved in the suppression of EMT via activation of TRPV1 channel/ In the future, we will aim to create less irritating generic cataract suppressors using capsaicin as a lead compounds.
