外層キトサンハイドロゲル、内層コラーゲンハイドロゲルで構成された人工神経(CCN)を作成した。シュワン細胞をコラーゲン層に封入しラット左坐骨神経欠損部に移植し(CCN+群)、CCN細胞なし群(CCN-群)、同細胞をシリコンチューブに封入した群(Silicone+群)、自家神経移植群(Auto群)と比較検討した。CCN+群はCCN-群およびSilicone+群と比較して有意な運動機能回復、髄鞘形成を示した。CCN-群とSilicone+群は歩行解析、組織学的解析いずれにおいても有意差を認めず、Auto群は最も優れた回復を示したが、再生軸索断面積に関しては CCN+群と有意差を認めなかった。 We prepared a chitosan-collagen hydrogel nerve conduit (CCN) consisting of an outer layer of chitosan hydrogel and an inner layer of collagen hydrogel to encapsulate the intended cells. Rats with a 10-mm sciatic nerve defect were treated with Schwann cells encapsulated in CCN (CCN+), CCN without SCs (CCN-), SC-encapsulated silicone tube (silicone+), and autologous nerve transplanting (auto). Behavioral and histological analyses indicated that motor function recovery and myelination of the CCN+ group were superior to those of the CCN- and silicone+ groups, whereas the CCN- and silicone+ groups showed no significant differences in the recovery of motor function and nerve restoration. In conclusion, SC-encapsulated CCN possessed synergistic effect on peripheral nerve regeneration, especially remyelination of host SCs. The comparable recovery in the CCN- and silicone+ groups suggests that the structure of CCN and/or the effect of chitosan metabolites positively impact nerve regeneration.
