BEV投与によるVEGF-A以外のその他の血管新生pathway のリガンドと受容体の挙動に関しては、Bev単回投与後は大きな変動は認めなかったが、BEVを複数回投与した耐性化検体では、特にEphrinA2, PlGFの亢進の可能性が示唆された。また、BEV未治療の初発悪性神経膠腫検体(NB)、BEV奏功時の腫瘍検体（EB）、及びBEV耐性時の腫瘍検体（RB）による細胞株による検討では、RB株は顕著な浸潤性を示し、脳組織内にびまん性に浸潤し、モデルマウスの生存曲線も短かった。一方で、EB株に関しては、RB株と比較すると浸潤性は劣り、ほぼNB株と同等程度であった。 The present study used glioblastoma tissues obtained at 3 different settings: tumors of initial resection (NB), tumors resected following Bev therapy (EB), and recurrent tumors after Bev therapy (RB). The paired samples of the initial and post-Bev recurrent tumors from the same patients were included. The amount of CD3+ and CD8+ T cell infiltration increased in the refractory Bev group compared with the naive Bev group. Both Foxp3+ regulatory T cells and CD163+ tumor-associated macrophages significantly decreased in the effective or refractory Bev group compared with the naive Bev group. In EB group, status of alternative angiogenic pathway including Angiopoietin1,2/bFGF/EphrinA2/PlGF was not significantly changed. In contrast, the status of EphrinA2 and PlGF tended to be increased in the RB group. We have established several cell lines from the tissues of NB, EB, and RB groups. Survival curve of the RB-derived xenografts tended to be shorter than other xenografts.
