Note (General): 骨芽細胞は骨に転移した癌細胞の増殖や生存に重要な役割を果たしている。癌細胞と骨芽細胞の相互作用に関しては多くの報告があるが、大部分は両者が分泌する液性因子を介したものであり、癌細胞と骨芽細胞が直接接触することによって起きる相互作用に関しては不明な点が多い。我々はこれまでに溶骨性増殖を示す前立腺癌細胞と骨芽細胞が接触することによって破骨細胞誘導が促進されることを明らかにしてきたが、今回は前立腺癌細胞と骨芽細胞の接触による骨芽細胞の遺伝子発現の変動を cDNA マイクロアレイによって解析した。その結果、不死化したヒト骨芽細胞がPC-3 ヒト前立腺癌細胞と接触することによって脂肪細胞の分化マーカーである FABP4 やPPARγの発現上昇がみられること、及びマウスの骨芽細胞とPC-3 細胞の接触培養によって骨芽細胞のALP 活性が低下することが明らかになった。以上より、成熟骨芽細胞と前立腺癌細胞の接触により骨芽細胞の分化抑制が起こる可能性が示唆された。成熟骨芽細胞骨は骨基質形成に重要な役割を果たすが、前立腺癌細胞はその機能を低下させ、骨を脆弱化することにより浸潤を容易にしている可能性がある。
