Note (General): 悪性胸膜中皮腫（malignant pleural mesothelioma: MPM）に高発現するポドプラニン分子に注目し、ラット抗ヒトポドプラニン抗体NZ-1およびそのヒト･キメラ抗体NZ-8のMPMに対する特異的免疫療法の可能性を検討した。MPM細胞株および腫瘍組織におけるポドプラニン抗原は、細胞株では73%(11/15)、腫瘍組織では92%(33/36)に発現を認めた。NZ-1およびNZ-8はポドプラニン発現MPM細胞に対して、高い抗体依存性細胞傷害活性（antibody-dependent cellular cytotoxicity: ADCC）を示した。エフェクター細胞の解析では、NK細胞にADCC活性を認めたが、単球はADCC活性を示さなかった。以上からNZ-1およびNZ-8のADCCにはNK細胞が重要であることが明らかとなった。NZ-8はヒト単核球をエフェクター細胞とした場合、NZ-1に比較して有意に高いADCC活性を示した。補体依存性細胞傷害活性（complement-dependent cytotoxicity: CDC）についても、NZ-8はNZ-1に比較して高い活性を示した。ACC-MESO-4細胞のSCIDマウス皮下移植系を用いた検討では、ヒトNK細胞とNZ-8を投与した群に有意に腫瘍増殖抑制効果が確認された。また、NZ-8は他の抗ポドプラニン抗体D2-40とは認識エピトープが異なっており、正常細胞よりMPMに発現するポドプラニン分子を認識しやすい特徴を有していた。以上から、ヒト・キメラ抗ポドプラニン抗体NZ-8を用いた臨床応用の可能性が示唆された。
