Note (General): 我々が作成したケモカインCXCL14/BRAK(BRAK)を過剰発現するトランスジェニック(BRAKTg)マウスと野生型(Wt)マウスを用いて化学発癌に対する応答と、悪性黒色腫細胞の実験的肺転移に対する応答を調べた。 BRAK Tgマウスの大腸癌の数はWtの1/10であり有意(P<0.001)に少なかった。また、悪性黒色腫の数を変えて注射すると、すべての細胞数においてBRAKTgマウスの方がWtマウスより肺への転移数が有意に少なく、かつ、生存率が高かった。さらに、注入腫瘍細胞数が減少するほど両者の生存率の差は大きくなり、注入細胞数3x103ではWtマウスの生存率が50％に対してTgでは100%であった。また、NK細胞活性の阻害によりTgマウス、 Wtマウスともに転移腫瘍数が激増し、両者における肺転移にNK細胞が関与している事が明らかにされた。健康人でもBRAKTgレベルの発現をしているヒトが存在したので、ケモカインCXCL14/BRAKは今後の副作用のない新しい癌の治療法開発のための有望な標的分子と考えられる。
