Note (General): 我々は膵癌細胞でglycogen synthase kinase (GSK) 3βの発現と活性が高く，膵癌細胞の生存と増殖にGSK 3βが必須であることを見出し、GSK3β阻害によるがん治療効果を細胞レベルと担がん動物モデルで実証し，GSK3βが膵癌の新しい治療標的であることを同定した。GSK 3βの機能解析により、GSK3βが誘導するシグナルがp53がん抑制分子経路，Rb細胞周期制御経路、細胞不死化経路に影響することを明らかにした。また、ヒト膵癌細胞移植動物において薬理学的GSK3β阻害剤の併用がTP53INP1 (tumor protein 53-induced nuclear protein 1)を介するDNA修復機構を制御して、ゲムシタビン(GEM)の抗腫瘍効果を相乗的に増強することを実証した (Shimasaki T, et al. J Gastroenterol 2012)。本研究では，とくに膵癌細胞の形態特性と運動能に着目してGSK3βの機能を多角的に解析した。膵癌培養細胞のスクラッチアッセイとトランスウェルアッセイにより、GSK3β阻害剤は膵癌細胞の遊走と浸潤を抑制した。また、GEMにより誘導される膵癌細胞の形態変化とFAK (focal adhesion kinase)/Rac1/MMP-2 (matrix metalloproteinase-2)機軸経路がGSK3β阻害剤により抑制された。これらの基礎的解析と並行して，GSK3β阻害作用を有する既存医薬品の適応外使用とGEMの併用による進行膵癌治療の医師主導型第Ⅰ/Ⅱ相臨床研究 (UMIN 000005095)を継続中である。
