中枢神経の再生可能なサカナの視神経切断モデルを使用し、再生初期に発現が上昇する遺伝子を同定し、細胞生存や神経突起伸長作用を確認し、これらを神経再生初期分子とした。この候補物質としてHSP70, レチノール結合蛋白プルプリンを同定した。これらに引き続いてレチノイン酸シグナル分子の発現が上昇した。これら再生分子を成熟ラット網膜に導入することにより、ラット視神経がin vivoで再生できた。
Regeneration-associated genes (RAGs) were cloned from axotomized zebrafish retina. HSP70 (a stress protein) and purpurin (a retinol-binding protein) were rapidly induced in the fish retina after optic nerve injury. HSP70 was involved in cell survival of injured retinal ganglion cells (RGCs), whereas purpurin was involved in neurite sprouting from injured RGCs. These RAGs could easily induce regrowth of axotomized optic nerve in adult rat.
