Note (General): 本研究は複数の超微少液性情報変化を同時に解析できる多機能バイオセンサを開発するため、その基礎研究として、(1)微小透析プローブによる生理活性物質の分離と高感度測定法の確立、(2)基質選択性酵素センサの開発、(3)水晶振動子マイクロバランスを利用した免疫センサの開発の各研究テーマについて検討し、以下の結果を得た。 I.微小透析プローブ (1)In vitroでのノリアドレナリン相対回収率は約20%であり、その検出限界は500fg/40μlであった。 (2)ノリアドレナリンの回収に関する基礎実験をラット内側前頭前野および脾臓で行い、十分満足のいく結果が得られた。 II.酵素センサ (1)本研究で作製したグルコースオキシダーゼ固定化センサは、基質選択性、感度及び濃度依存性に関して十分満足のいくものであった。 (2)低溶存酸素溶液中では、グルコースセンサの感度の低下が認められるが、フェロセンを電子メディエータとして介在させることによりセンサの感度が維持できた。 III.免疫センサ (1)水晶振動子をマイクロバランスとして用いることができる。またその感度は1Hz/ngときわめて高い。 (2)水晶振動子電極表面にチラミンを電解重合し、アミノ基を導入後、グルタルアルデヒドを用い抗体(抗ヒトIgG)のアミノ基との間にシッフ塩基形成反応を行わせた。これにより抗体を電極表面に効率よく固定化できる。 (3)各種濃度のヒトIgG溶液(10,30,50μg/ml)投与によってきわめてゆっくりとした時間変化で振動数が変化し、IgG50μg/ml投与の場合、約20分後に最大振動数変化200Hzに達した。一方、ウシ血清アルブミン(10μg/ml)を投与しても、IgG投与時に観察されたような顕著な振動数変化が認められないことから、この反応は非特異的タンパク結合などによるものではない。
The purpose of this research project was to accomplish the biosensors which can detect the endogenous substances influencing neuronal activity in the brain. For this purpose, the characteristics of the following sensors were studied ; (1) microdialysis sensor for separation and analysis of bioactive substances, (2) enzyme biosensor for detection of glucose immobilized with glucose oxidase and (3) a quartz crystal microbalance sensor for immunosorbent assay.1. Microdialysis biosensorNoradrenaline release in the medial prefrontal cortex and spleen of rats was studied. The detection limit of the microdialysis sensor system for noradrenaline was 500 fg/40 mul at a 3 : 1 signal to noise ratio. A series of experiments using pharmacological and physiological manipulations indicated that the noradrenaline recovered in the dialysate was mainly derived from nerve terminals.2. Enzyme biosensorGlucose oxidase was immobilized onto platinum electrode to attain stabilized activity. The concentration response characteristics and the stability of the sensor were studied for glucose concentrations of 0.1-10.0mM.Steady state response of the sensor was proportional to the glucose concentration up to 5 mM.The slope of the linear correlation of sensor response was inversely proportional to the dissolved oxygen concentration. The decrease in sensor response caused by the reduction of dissolved oxygen concentration was overcome by using ferrocene carboxialdehyde as an electron mediator.3. Quartz crystal microbalance sensorDetermination of human immunoglobulin G (IgG) was performed by using a piezoelectric crystal biosensor system, which was consist of an AT-cut quarts piezoelectriccrystal, oscillator and frequency counter. Anti-human IgG was immobilized onto the surface of the gold electrode with tyramine and glutaraldehyde. The resonant frequency shift resulting from the affinity reaction of anti-IgG and IgG correlated with a concentration of IgG in the range of 10mug/ml-50mug/ml. The crystal sensor showed no response to bovine serum albumin.
