レトロウイルス挿入変異によってマウスに発症した腫瘍から、ウイルスタギングを用いて新しいがん関連因子群の探索を進め、これまでにヒストンやDNAのメチル化の制御に関与する酵素群を同定した。さらに、これらのエピジェネティクス制御因子が、がんの発症ばかりでなく、がんの転移、薬剤耐性など悪性進展の様々なステップにおいて重要な役割を担っていることを示す新しい知見を得ることができた。
Retroviral insertional mutagenesis in mice is one of the powerful strategies for high-throughput identification of cancer genes. Using this system, we have frequently identified the genes encoding the enzymes engaged in histone or DNA methylation. We have found that some of the enzymes are involved not only in the tumor initiation but also in the malignant tumor progression such as cell invasion and drug resistance.
