機能的に正常な免疫システムを維持するうえで、T細胞を中心とした自己の正常免疫組織・細胞による、非自己である病原性微生物やがん細胞の識別過程を理解することが必要不可欠である。本研究では、免疫学的役割が明らかではない胸腺樹状細胞サブセット(Sirpα^樹状細胞)に着目し、詳細な解析を行った。その結果、胸腺内Sirpα+樹状細胞による、血液中のタンパク抗原を標的とした新規の免疫寛容誘導経路を同定した。また、この経路を介して、分泌性の腫瘍抗原に対して抗腫瘍免疫寛容が誘導されることも明らかにした。
The discrimination of self or non-self antigens is essential for the maintenance of intact immune system. In this study, we demonstrated that a thymic dendritic cell (DC) subset, Sirpα+ DC is the major conductor of the novel process of intrathymic immune tolerance against blood-borne antigens. Moreover, the secretory tumor-specific antigens can induce the intrathymic anti-tumor immune tolerance through the antigen presentation by thymic Sirpα+ DCs.
