がん抑制遺伝子RBは多くのクロマチン修飾因子と結合し、その機能を制御する。このようなRBによるエピジェネティック制御機能を解明するために、我々はRB欠損モデルマウスとヒト原発がん組織を使い、RBとATMの相互作用がDNAメチル化酵素DNMT1の安定性を制御すること、また、DNAマイクロアレイ解析から数百のがん関連遺伝子のDNAメチル化状態が、RB-ATM-DNMT1の相互作用により調節されていることを発見した。
The retinoblastoma tumor suppressor gene (RB) product has been implicated in epigenetic control of gene expression due to its ability to physically bind to and regulates many of chromatin modifying factors. To elucidate the mechanism of RBepigenetic function, we performed genetic and epigenetic analyses in an Rb-deficient mouse and human primary cancer tissues. Her e we report that the genetic interaction of Rb and ATM regulates DNMT1 protein stability, and microarray analysis indicated that RB-ATM-DNMT1 interaction controls the DNA methylation status in the promoter of hundreds of cancer related genes.
