B 型肝癌組織から樹立した新規肝癌細胞KM 細胞を用いてHBV 陽性細胞で発現上昇する2 遺伝子を同定した。その2 遺伝子の発現抑制によりcccDNA を抑制し、エンテカビルとの併用で95%以上のcccDNA の抑制を認めた。NTCP発現HepG2細胞で2 遺伝子をノックアウトした細胞、キメラマウス由来初代肝細胞（PXB）、及びレンチウイルス発現系にて2 遺伝子を過剰発現した細胞を樹立しHBVの複製を評価した。2 遺伝子のノックアウト細胞ではHBV-DNA、cccDNA、pgRNAの低下、過剰発現細胞ではそれらの増加を認めた。
We identified two essential host genes for HBV replication by using single cell transcriptome analysis of KM cells that were established from a HCC tissue derived from a patient with chronic hepatitis B. Repression of two genes reduced cccDNA, and by the combination of ETV, more than 95% reduction of cccDNA was obtained. By using HepG2-NTCP cells in which two genes were knocked down, primary human hepatocytes (PXB) and HepG2-NTCP cells in which two genes were overexpressed by lentivirus system, we showed HBV-DNA, cccDNA and pgRNA were substantially changed by the expression of two genes.
