テロメア伸長酵素として知られるテロメレースの触媒サブユニットであるhTERTは、悪性腫瘍に特異的に高発現する蛋白である。近年、TERTがテロメア伸長以外の機能を有することが示唆されている。我々は、hTERTがRNA分子であるRMRPと結合して、RNA依存性RNAポリメレース(RdRP)活性を示すことを見出した。RMRPは正常細胞株から癌細胞株まで幅広く発現している。hTERTとRMRPとを共に発現している子宮癌・乳癌細胞株を用いた検討では、hTERT-RMRP複合体のRdRP活性により、細胞内でRMRPを鋳型にした長い2本鎖RNAの合成、さらには2本鎖RNAの切断によってRMRPに由来するsmall RNAが生成されることがわかった。このRMRP由来small RNAは塩基配列特異的に遺伝子発現を抑制すること示唆された。
hTERT elongates telomere through its telomere-specific RNA dependent DNA polymerase activity. Accumulating evidence indicates hTERT has other function(s) beyond telomere maintenance. We identified RMRP as a novel binding partner of hTERT. hTERT-RMRP complex exhibited RNA dependent RNA polymerase (RdRP) activity and synthesized complementary strand of RMRP in vivo. The short RNA derived from the product of the RdRP activity was supposed to be involved in RNA silencing.
