Note (General): 3-エトキシシクロブタノン誘導体を適切なルイス酸を用いて活性化して1, 4-双極性活性種を生成させ、アリルシラン、シリルエノールエーテル、イミン、およびインドール類と反応させたところ、目的とする形式的[ 4+2]環化付加反応が円滑に進行し、対応する六員環化合物が効率的に得られることを明らかにした。さらにこの反応を用いて、生理活性物質であるブレマゾシンおよびアスピドスペルジミンの全合成を達成した。
We found that formal[ 4+2] cycloaddition reactions of 3-ethoxycyclobutanones with allylsilanes, silyl enol ethers, imines, or indoles proceeded smoothly to afford the corresponding six-membered cyclic compounds in high yields by activating the cyclobutanones with appropriate Lewis acids. We accomplished total synthesis of(±)-bremazocine and(±)-aspidospermidine by using the new cycloaddition.
