アレルギー性疾患の原因については、胎児期あるいは小児期の前半に最も頻繁に感染するRespiratory syncytialウイルスなどのウイルスの関与がよく知られるようになったが、その病態の背景である感受性遺伝子との相互作用はよく知られていない。妊娠モデルマウスの児の脾臓細胞のIL4RA遺伝子におけるCpG部位のメチル化と感染の関係が出産直後において認められた。疫学研究では、臍帯血単核球のIL4RAにおけるCpG部位のメチル化がハウスダスト感作による喘息との関係において認められた。これらの結果から、出生直後の児において感染が成立するとともにアレルギー関連の遺伝子が発現していると考えられた。
The infection of virus such as respiratory syncytial virus is involved in pathogenesis of allergy during prenatal and postnatal infants, although the interaction between candidate genes and virus infection remains to be elucidated. We demonstrated hypomethylation of CpG islands produced by virus infection in IL4RA gene in splenic cells for postpartum infants from pregnant model mouse. Epidemiologic study showed the hypomethylation of CpG islands in in IL4RA gene in monocytes in cord blood of pregnant patients with asthma sensitized by house dust. These results suggest expression of IL4RA gene in just new born infants is produced by infection of RS virus.
