心不全の病態形成における解析を行うために、マウスを用いて圧負荷誘導性の心肥大・心不全モデルの作成を行った。C57/B6マウスに、人工呼吸器管理下に横行大動脈に狭窄を作成した。術後2週で有意な左室肥大が、術後4週で肺うっ血を生じ心不全が起こることを確認した。筋特異的ユビキチン連結酵素MAFbxが、術後1週、2週と時間依存的にその発現レベルが有意に増加していくことを確認した。MAFbx欠損マウスに圧負荷モデルを作成し、MAFbx欠損マウスでは圧負荷による左室肥大は有意に抑制された。MAFbxは圧負荷に伴う心肥大および心臓リモデリングを制御している可能性が示唆された。
