本研究では、歯髄炎治療薬としてのMMP-3の作用メカニズム解明のために、ヒト歯髄組織中からMMP-3結合タンパクを検索した。解析結果から、バーシカンをMMP-3の基質の1つとして同定した。イヌ歯髄炎モデルにおいて、MMP-3処理した歯髄組織は対照群と比較してバーシカンの局在と構造が異なっていた。さらに、切断されたバーシカンとヒアルロン酸の複合体は炎症細胞の結合能力が著しく低下していた。以上から、バーシカンとヒアルロン酸の複合体は炎症細胞の集積に重要であり、MMP-3による抗炎症効果はバーシカン切断によるヒアルロン酸複合体形成阻害によるものと考えられた。
In this study, to clear the mechanism of the effect of MMP-3 as anti-pulpitis drugs, we identified the MMP-3 binding protein in human dental pulp tissues. From results of analyses, versican was identified as substrate of MMP-3. On canine pulpitis model, structure and localization of versican on MMP-3 treated pulp tissue was different to control non treated tissues. In conclusion, complex of versican and hyaluronic acid was critical in recruiting of inflammatory cells, and anti-inflammatory effects of MMP-3 were closely linked to disruption of versican and hyaluronic acid.
