申請者はこれまでに、肝由来分泌タンパクセレノプロテインP(SeP)がインスリン抵抗性を誘導して高血糖を発症させるヘパトカインであることを報告した。本研究では、SePによる運動療法抵抗性の発症という新たな病態概念を提唱した。筋管細胞において、SePが活性酸素依存性AMPKリン酸化を減弱すること、SeP欠損マウスの運動感受性が亢進していること、SePの筋受容体がLRP1であることを見出した。SePの過剰は受容体LRP1を介して筋で活性酸素/AMPK経路を抑制することで“運動抵抗性”を惹起する。SeP/LRP1の阻害薬は運動療法の感受性を増強させる新たな2型糖尿病治療薬となる可能性がある。
We have previously identified selenoprotein P (SeP) as a type 2 diabetes-upregulated hepatokine that induces insulin resistance and hyperglycemia. In this study, we propose a novel pathological concept SeP-induced exercise resistance. We have found out that SeP impairs ROS-induced AMPK phosphorylation in cultured myotubes. SeP knockout mice showed increased sensitivity to exercise training. siRNA screening revealed that LRP1 functions as a SeP receptor in cultured myotubes. This study demonstrates that overproduction of SeP induces exercise resistance by suppressing ROS/AMPK through muscle receptor LRP1. Our study opens the door for inhibitors of SeP/LRP1 axis as new drugs for physical inactivity-associated diseases such as type 2 diabetes.
