本研究においては、ウサギ動物実験モデルに対する大腿動脈からのカテーテルを用いたアプローチ法を確立した。次に、細径カテーテルを用いる事により肝動脈の可及的末梢の選択を行う事に成功した。肝臓を標的とした血管バリアー機能の評価を行うために、定常状態での薬剤投与による影響を評価したが、NOドナーであるニトログリセリン、SNP、また血管透過性を亢進させるヒスタミン、PAFの投与下における血管透過性の評価をエバンスブルーを用いて行ったが、非癌肝では変化は認めなかった。一方、VX2腫瘍モデルにおいては、腫瘍周囲肝実質において、ニトログリセリン投与下で血管透過性の著明な亢進が確認できた。
In this project, we obtained the protocol of noninvasive intravascular interventional approach from femoral artery and catheterization to selective hepatic arteries. In steady condition, there is no sign of extravasation of FITC-dextran injected from hepatic artery. And there is no sign of extravasation of FITX-dextran in the drug administration model of nitroglycerin, sodium nitroprusside, histamine, and platelet activating factor. On the other hand, in liver VX-2 tumor model, the hyper-vascular permeability was observed at the peritumoral area in administration of nitroglycerin.
